Article Abstract

Carcinoembryonic antigen (CEA) isolated from metastases of colon adenocarcinoma was subjected to deglycosylation with liquid hydrogen fluoride. The protein fraction obtained (PF CEA) was used for to prepare monospecific antiserum to PF CEA. Comparative studies of CEA, PF CEA, and non-specific cross-reacting antigen (NCA-1) have been carried out using monospecific antisera. Circular dichroism spectra of CEA and PF CEA have been studied. The data obtained suggest that some immunodominant regions of CEA are topographic, and their formation needs a specific conformation of the macromolecule, which is stabilized by the sugar moiety.

In the ancient microbial Wood-Ljungdahl pathway, carbon dioxide (CO) is fixed in a multistep process that ends with acetyl-coenzyme A (acetyl-CoA) synthesis at the bifunctional carbon monoxide dehydrogenase/acetyl-CoA synthase complex (CODH/ACS). In this work, we present structural snapshots of the CODH/ACS from the gas-converting acetogen , characterizing the molecular choreography of the overall reaction, including electron transfer to the CODH for CO reduction, methyl transfer from the corrinoid iron-sulfur protein (CoFeSP) partner to the ACS active site, and acetyl-CoA production. Unlike CODH, the multidomain ACS undergoes large conformational changes to form an internal connection to the CODH active site, accommodate the CoFeSP for methyl transfer, and protect the reaction intermediates.

Ni(II) complexes with an integer spin = 1 that behave as clock transition spin qubits at zero magnetic field are resilient to magnetic fluctuations of the spin bath, while Co(II) complexes with a half-integer spin ( = 3/2) lose their coherence when they are subject to the same fluctuating magnetic field as the Ni(II) ones. These findings demonstrate that adequately designed Ni(II) complexes are excellent candidates for spin qubits.
