Drosophila melanogaster is one of the most widely used genetic model systems in biology. The ease of working in an invertebrate model system allows the design and execution of many experiments that would be infeasible in a vertebrate model. Although the strength of the fly as a model system lies primarily in the ease of genetic manipulation, it is flexible enough that biochemical and proteomic approaches can also be used to build a more comprehensive study. Here we present a pair of complementary protocols that we have used to examine sirtuin biology in Drosophila. First, we describe our protocol for measuring lifespan in flies expressing a gene of interest under the inducible control of the Gene-Switch system. Finally, we describe a method for performing chromatin immunoprecipitation on adult flies, including some of the difficulties associated with using this technique in chitinous tissue.

Mitochondrial biogenesis requires the expression of genes encoded by both the nuclear and mitochondrial genomes. However, aside from a handful transcription factors regulating specific subsets of mitochondrial genes, the overall architecture of the transcriptional control of mitochondrial biogenesis remains to be elucidated. Short-chain enoyl-CoA hydratase 1 deficiency (ECHS1D) is a rare genetic disorder caused by biallelic pathogenic variants in the ECHS1 gene. ECHS1D is characterised by severe neurological and physical impairment that often leads to childhood mortality. Therapies such as protein and single nutrient-restricted diets show poor efficacy, whereas the development of new treatments is hindered by the low prevalence of the disorder and a lack of model systems for treatment testing.

Biocontrol techniques that impair reproductive capacity of insect pests provide opportunities to control the dynamics of their populations while minimizing collateral damage to non-target species and the environment. The Trojan Female Technique, or TFT, is a method of the trans-generational fertility-based population control through the release of females that carry mitochondrial DNA mutations that negatively affect male, but not female, reproductive output. TFT is based on the evolutionary hypothesis that, due to maternal inheritance of mitochondria, mutations which are beneficial or neutral in females but harmful in males can accumulate in the mitochondrial genome without selection acting against them.
