Targeted Alpha Therapy has shown great promise in cancer treatment, sparking significant interest over recent decades. However, its broad adoption has been impeded by the scarcity of alpha-emitters and the complexities related to their use. In 2002, in the Emilia-Romagna region of Italy, a comprehensive strategic plan was developed with the aim of improving the integration and efficiency of the genetic services. Two decades later, this report aims to explore the current functioning of the regional network, with special focus on clinical genetics in the evolving scenarios. To this aim, we analyzed the activity data of the medical genetics services in the region, to identify and possibly improve currently open issues.

The protein therapeutics market, including antibody and fusion proteins, has experienced steady growth over the past decade, underscoring the importance of optimizing amino acid sequences. In our previous study, we developed a fusion protein, R31, which combines retinol-binding protein (RBP) with albumin domains IIIA and IB, linked by a sequence (AAAA), and includes an additional disulfide bond (N227C-V254C) in IIIA. This fusion protein effectively inhibited hepatic stellate cell activation.

The rapidly growing glycoscience has boosted the research on the synthesis of glycans and their conjugates, which are centered on the stereoselective formation of glycosidic bonds. Compared to the mainstream acid-promoted glycosylation method that undergoes the S1 type mechanism, the basic/neutral conditions give better stereo control via the S2 mechanism. Anomeric hydroxyl group transformation, whether to form glycosidic bonds directly or to install a leaving group for later glycosylation, is key to carbohydrate synthesis, and the strategies in the stereo control of these reactions under basic/neutral conditions are summarized in this review.
