Exposure to high sustained positive acceleration (+Gz) is known to have a pathophysiological effect on the heart of the rat. As critical regulators of cardiac myocyte survival and death, mitochondria may be crucially involved in +Gz-induced pathogenesis. It was, therefore, of interest to investigate myocardial mitochondrial ultrastructure, respiratory function, and antioxidant capacity in rats after exposure to +10 Gz for 5 min. The results showed that high +Gz stress could damage mitochondrial ultrastructure; this was apparent from swollen, degenerated, and reduced mitochondria, and mitochondrial cristae broken or disappeared. This resulted in significant changes of quantitative indicators of mitochondria morphometry, for example increased surface density, volume density, average volume, and average surface area, and reduced numerical density. The studies also revealed that exposure to +Gz stress induced dysfunction of the mitochondrial respiratory chain, reduced the activity of antioxidant enzymes (catalase, superoxide dismutase, and glutathione peroxidase), and increased malondialdehyde content. We thus conclude that high +Gz stress not only damaged mitochondrial ultrastructure but also impaired respiratory function and antioxidant capacity.

Post-streptococcal autoimmune neuropsychiatric disorders (PANDAS) are a group of pathological condition characterized by sudden-onset obsessive-compulsive and tic disorders following beta-hemolytic Streptococcus group A (GAS) infection, hypothesized to be caused by autoimmune mechanisms targeting the basal ganglia. Scant literature is available regarding the microbiota composition in children with PANDAS, however few studies support the hypothesis that streptococcal infections may alter gut microbiota composition in these patients, leading to chronic inflammation that may impact the brain function and behavior. Notable changes include reduced microbial diversity and shifts in bacterial populations, which affect metabolic functions crucial for neuroinflammation.

CAR-NK therapy is becoming a promising approach to treat solid tumors. However, the random insertion of the CAR gene and inflexible CAR expression caused by common preparation methods significantly impact its efficacy and safety. Here we successfully established a novel type of CAR-NK cells by integrating CAR sequences into the GAPDH 3'UTR locus of NK-92MI cells (CRISPR-CAR-NK), achieving site-specific integration of the CAR gene and allowing endogenous regulatory components to govern CAR expression.

Chronic obstructive pulmonary disease (COPD) is a progressive lung disease characterized by obstructed airflow, airway remodeling, and inflammation, with cigarette smoke (CS) exposure being the main risk factor. While CS extract (CSE) has been shown to activate caspases in various cell types, the role of caspases in human lung fibroblasts (hLFs), in COPD remains poorly understood. Recent studies have linked caspases to extracellular matrix (ECM) remodeling in skin and kidney fibrosis.

Nanozymes with multienzyme-like activity have sparked significant interest in anti-tumor therapy via responding to the tumor microenvironment (TME). However, the consequent induction of protective autophagy substantially compromises the therapeutic efficacy. Here, a targeted nanozyme system (Fe-Arg-CDs@ZIF-8/HAD, FZH) is shown, which enhances synergistic anti-tumor ferroptosis/apoptosis therapy by leveraging machine learning (ML).

Human musicality might have co-evolved with social cognition abilities, but common neuroanatomical substrates remain largely unclear. In behavioural variant frontotemporal dementia, social cognitive abilities are profoundly impaired, whereas these are typically spared in Alzheimer's disease. If musicality indeed shares a neuroanatomical basis with social cognition, it could be hypothesized that clinical and neuroanatomical associations of musicality and social cognition should differ between these causes of dementia.
