It is demonstrated how acyclic diene metathesis polymerization (ADMET) provides an efficient strategy for the labeling of polyolefins. The versatility of phosphorus chemistry allows designing substituted BODIPY monomers or chain stoppers for the synthesis of precise labeled (degradable) polyphosphoesters.

A concise synthetic route to an epimer of the recently isolated biologically active cyclic tetrapeptide koshidacin B has been developed, which featured a late-stage functionalization of a macrocyclic scaffold through a cross metathesis reaction. The synthetic 9--koshidacin B showed marginal differences in spectroscopic behavior with that of the natural product but exhibited conformational preferences similar to those reported for analogous substrate chlamydocin. Moreover, it exhibited a useful level of selective inhibition of biologically relevant enzyme histone deacetylase 1 with an IC value of 0.

Sb-to-P Metathesis: A Direct Route to Structurally Constrained, Cationic P Compound.

Angew Chem Int Ed Engl

November 2024

School of Chemistry, Raymond and Beverly Sackler Faculty of Exact Sciences, Tel Aviv University, 69978, Tel Aviv, Israel.

Structurally constrained, cationic P compound [LP][SbCl] with an OCO pincer-type ligand (L) having a central carbene donor was directly synthesized via an Sb-to-P metathesis reaction between PCl and LSb-Cl. [LP][SbCl] was isolated and its reactivity with small molecules (ROH and RNH) was studied, showing that [SbCl] is not an innocent counter anion, but an active participant in these reactions. When the [SbCl] was replaced with the [CBH] ([Cb]) anion, the reactions were redirected to [LP] cation only.

A new and highly efficient ring-closing metathesis-based strategy was developed for the synthesis of the cyclic urea 1,3-diazepinone, presenting significant improvement upon previous methods. Using a direct glycosylation approach, analogues of the potent cytidine deaminase (CDA) inhibitor diazepinone riboside were then synthesized including 2'-deoxyribo-, 2'-deoxy-2'-fluoroarabino-, and 2'-deoxy-2',2'-difluoro-diazepinone nucleosides, all with moderate to good yield and excellent anomeric selectivity. Crucially, in contrast to the previous multistep linear synthesis of 2'-deoxyribo- and 2'-deoxy-2'-fluoroarabino-diazepinone nucleosides, this is the first report of direct glycosylation to access these nucleosides.

New synthetic routes have been developed to synthesize osmaborane clusters featuring B and B rings in the coordination spheres of osmium. Thermolysis of [Os(PPh)Cl], in the presence of excess of [BH·THF] led to the formation of [Os(PPh)H(η-BH)], along with [HOs(PPh)BH], . Cluster features a planar tetraborane ring coordinated to an osmium center in an η fashion.

A new activity-based probe (ABP) of cysteine proteases (FGA139) has been designed and synthesized. The design of the ABP has been done based upon the chemical structure of an irreversible inhibitor of cysteine proteases by attaching a bodipy fluorophore at the N-terminus of the peptide backbone. The synthetic route of the probe has a metathesis and a "click" reaction as key steps.
