Stentorin serves as the photoreceptor for the photophobic and negative phototactic responses in Stentor coeruleus. Two forms of the stentorin have been isolated and purified. The strongly fluorescent form, stentorin I at pH 7.8, exhibited nearly exponential fluorescence decay monitored at 620 nm, having two comparable lifetime decay components of 2.53 ns (47%) and 5.95 ns (53%). Stentorin I showed no significant time-resolved fluorescence emission spectra in the picosecond-nanosecond time scales. The weakly fluorescent form, stentorin II, exhibited an ultrafast fluorescence decay component (10 ps) at an emission wavelength of 630 nm and pH 7.8. The amplitudes of the multi-component fluorescence in stentorin II were found to be emission wavelength-dependent. Furthermore, the fluorescence emission spectrum was time-resolvable in the picosecond time scales. Effects of pH and pD on the fluorescence decay kinetics and time-resolved spectra of stentorins I and II have also been investigated. Results are suggestive of proton dissociation as a primary photoprocess from the excited state of stentorin II.

Live imaging techniques have revolutionized our understanding of paracrine signaling, a crucial form of cell-to-cell communication in biological processes. This review examines recent advances in visualizing and tracking paracrine factors through four key stages: secretion from producing cells, diffusion through extracellular space, binding to target cells, and activation of intracellular signaling within target cells. Paracrine factor secretion can be directly visualized by fluorescent protein tagging to ligand, or indirectly by visualizing the cleavage of the transmembrane pro-ligands or plasma membrane fusion of endosomes comprising the paracrine factors.

N-heterocyclic carbene (NHC)-protected gold nanoclusters display high stability and high photoluminescence, making them well-suited for fluorescence imaging and photodynamic therapeutic applications. We report herein the synthesis of two bisNHC-protected Au nanoclusters with π-extended aromatic systems. Depending on the position of the π-extended aromatic system, changes to the structure of the ligand shell in the cluster are observed, with the ability to correlate increases in rigidity with increases in fluorescence quantum yield.

Chromophores incorporated into rigid polymer matrices may exhibit novel photophysical properties distinct from those in liquid solutions. In this work, we explored the decay path of the second ππ* state (2ππ*) of riboflavin in poly(vinyl alcohol) (PVA) solutions and films with various acidities. Highly efficient internal conversion from 2ππ* to the lowest ππ* state (1ππ*) induced by slight in-plane motion is demonstrated in all PVA solutions and films, irrespective of environmental acidity and rigidification.
