Here we report the draft genome sequence of Bacillus psychrosaccharolyticus, a cold-adapted bacterium with biotechnological interest. The genome contains genes related to the ability of this microorganism to grow at low temperatures and includes a nucleoside 2'-deoxyribosyltransferase, which can be used in the industrial synthesis of modified nucleosides with therapeutic activity.

The synthesis of nucleosides is crucial for pharmaceutical and biotechnological applications, acting as drugs and as essential building blocks for numerous therapeutic agents. However, most enzymes employed in nucleoside biocatalysis are not recycled, possess limited stability, and have strict substrate selection for ribonucleosides or 2'deoxyribonucleosides. We employed 2'-deoxyribonucleoside transferase (NDT) enzymes from thermophilic and psychrophilic bacteria to demonstrate they can be immobilized to enhance specific activity, stability, and recyclability.

Nucleoside 2'-deoxyribosyltransferase (NDT) from the psychrophilic bacterium Bacillus psychrosaccharolyticus CECT 4074 has been cloned and produced for the first time. A preliminary characterization of the recombinant protein indicates that the enzyme is an NDT type II since it catalyzes the transfer of 2'-deoxyribose between purines and pyrimidines. The enzyme (BpNDT) displays a high activity and stability in a broad range of pH and temperature.

Processes for the biotechnological production of kerosene and diesel blendstocks are often economically unattractive due to low yields and product titers. Recently, Clostridium acetobutylicum fermentation products acetone, butanol, and ethanol (ABE) were shown to serve as precursors for catalytic upgrading to higher chain-length molecules that can be used as fuel substitutes.
