A new conjugated phosphonium salt TPP was synthesized readily from phosphine-triggered ring-opening of 2,4,5-triphenylpyrylium salt. In aqueous solution, it exhibited interesting AIEE properties and self-assembled into fluorescent nanoparticles, which can be used as a fluorescence probe to image mitochondria in cells.

Mitochondria are the seat of cellular energy and play key roles in regulating several cellular processes such as oxidative phosphorylation, respiration, calcium homeostasis and apoptotic pathways. Mitochondrial dysfunction results in error in oxidative phosphorylation, redox imbalance, mitochondrial DNA mutations, and disturbances in mitochondrial dynamics, all of which can lead to several metabolic and degenerative diseases. A plethora of studies have provided evidence for the involvement of mitochondrial dysfunction in the pathogenesis of neurodegenerative diseases such as Parkinson's disease, Alzheimer's disease, Huntington's disease, and amyotrophic lateral sclerosis.

Anticancer activity of salinomycin quaternary phosphonium salts.

Eur J Med Chem

January 2025

Department of Medical Chemistry, Faculty of Chemistry, Adam Mickiewicz University, Uniwersytetu Poznańskiego 8, 61‒614, Poznań, Poland. Electronic address:

In recent years salinomycin has emerged as a promising anticancer drug. Many literature reports have proved its remarkable antiproliferative activity. Moreover, chemical modifications of salinomycin lead to analogues with even higher cytotoxicity against cancer cell lines and a better selectivity index for malignant cells than those of the unmodified compound or a standard anticancer drug such as doxorubicin.

Mitochondria are considered as promising targets for cancer treatment. In the present study, triphenyl phosphonium cationic group-conjugated fisetin (mito-fisetin) was synthesized, and its anticancer activity was investigated in several cellular models of estrogen receptor (ER)-positive breast cancer in vitro and in vivo in proliferating and tamoxifen-promoted senescent states. Mito-fisetin, when used at low micromolar concentrations, stimulated the dissipation of mitochondrial membrane potential and oxidative stress, and affected mitochondrial function, resulting in apoptosis induction in senescent breast cancer cells.

Article Synopsis
  • Heteropolycyclic molecular systems, key in materials and pharmacology, often involve 2D aromatic rings, but this study introduces a new 3D hybrid structure by combining an aromatic boron cluster with a phosphine and a π-conjugated unit.
  • A novel synthetic approach utilizing B-H activation of nido-carboranes with alkynes results in twisted boron cluster-extended phosphoniums, achieving high yields under mild conditions.
  • These new 3D structures show enhanced properties like improved thermal and chemical stability, as well as better photophysical characteristics, highlighting their potential applications in functional materials.

Muconic acid is a six-carbon dicarboxylic acid with conjugated double bonds that finds extensive use in the food (additive), chemical (production of adipic acid, monomer for functional resins and bio-plastics), and pharmaceutical sectors. The biosynthesis of muconic acid has been the subject of recent industrial and scientific attention. However, because of its low concentration in aqueous solutions and high purity requirement, downstream separation presents a significant problem.
