Cytotoxicity of a compound is determined by the intracellular concentration mediated both by passive permeability and active uptake through drug transporters. However, the major liver uptake transporters were either absent or expressed at significantly lower levels in human liver cell lines than in human liver. When comparing cytotoxicity of five statins, the organic anion transporting polypeptide 1B1 expressing HEK cells showed a significantly higher sensitivity than the wild-type HEK cells. The IC50 shifts ranged from 9- to >100-fold, and the potency shifts collapsed in the presence of rifampicin, the inhibitor for OATPs. The extent of the IC50 shift correlated with the permeability of the statins with high permeable compounds having smaller shifts and low permeable compounds having larger shifts. The changes in statin potency in transporter-transfected cells reflect the active uptake of statins into the cells, and the increased intracellular drug concentration lead to increased toxicity. The data suggested that uptake transporters have a significant impact on the outcomes of a cell-based assay and should be considered during the early stages of compound toxicity screening in drug discovery. For compounds with low permeability that are likely to undergo transporter-mediated uptake, it is important to test them in transporter-competent cell models.

Compared with Zn, the current mainly reported charge carrier for zinc hybrid capacitors, small-hydrated-sized and light-weight NH is expected as a better one to mediate cathodic interfacial electrochemical behaviors, yet has not been unraveled. Here we propose an NH-modulated cationic solvation strategy to optimize cathodic spatial charge distribution and achieve dynamic Zn/NH co-storage for boosting Zinc hybrid capacitors. Owing to the hierarchical cationic solvated structure in hybrid Zn(CFSO)-NHCFSO electrolyte, high-reactive Zn and small-hydrate-sized NH(HO) induce cathodic interfacial Helmholtz plane reconfiguration, thus effectively enhancing the spatial charge density to activate 20% capacity enhancement.

The response of oilseed rape to sulfur (S) restriction usually consists of increasing the components of S utilization efficiency (absorption, assimilation and remobilization) to provide S to seeds. However, source-sink relationships and S management in developing seeds under sulfate restriction are poorly understood. To address this, impacts of sulfate restrictions applied at "visible bud" or "start of pod filling" stages were studied with two genotypes (Aviso, Capitol) showing similar seed yield but higher seed weight and lower number of seeds per plant for Capitol under non-limited conditions.

Purpose Of Review: Patients that present with a physical frail phenotype have a higher risk of poor kidney transplant outcomes and are therefore less likely to be wait listed for a transplant. The physical frailty phonotype is more prevalent in older adults >65years with chronic and end stage kidney disease, thus partly contributing to inequitable access to transplant. Frailty can potentially be reversed by prehabilitation.
