In the cell nucleus, chromosomes have a complex spatial organization, spanning several length scales, which serves vital functional purposes. It is unknown, however, how their three-dimensional architecture is orchestrated. In the present article, we review the application of a model based on classical polymer physics, the strings and binders switch model, to explain the molecular mechanisms of chromatin self-organization. We explore the scenario where chromatin architecture is shaped and regulated by the interactions of chromosomes with diffusing DNA-binding factors via thermodynamics mechanisms and compare it with available experimental data.

Background: The homologous recombination deficiency (HRD) test is an important tool for identifying patients with epithelial ovarian cancer (EOC) benefit from the treatment with poly(adenosine diphosphate-ribose) polymerase inhibitor (PARPi). Using whole exome sequencing (WES)-based platform can provide information of gene mutations and HRD score; however, the clinical value of WES-based HRD test was less validated in EOC.
Methods: We enrolled 40 patients with EOC in the training cohort and 23 in the validation cohort.

Mass controls two major processes in volcanic islands: large-scale collapse and vertical movements. Therefore, large islands like Hawaii are gradually subsiding and have undergone massive landsliding. What if the mass is much smaller, and there is good evidence that the vertical movement is more complex than simple loading-related subsidence? Here, we show that small volcanic islands, seemingly stable because of the small mass, can undergo sector collapses that can affect the vertical movement of the island.

Ultrafine particles (UFPs) under 100 nm pose significant health risks inadequately addressed by traditional mass-based metrics. The WHO emphasizes particle number concentration (PNC) for assessing UFP exposure, but large-scale evaluations remain scarce. In this study, we developed a stacking-based machine learning framework integrating data-driven and physical-chemical models for a national-scale UFP exposure assessment at 1 km spatial and 1-hour temporal resolutions, leveraging long-term standardized PNC measurements in Switzerland.
