1. The kinetics and inhibition of p-nitrophenylacetate hydrolysis by cytosolic esterases of female alfalfa leafcutting bees, Megachile rotundata (Fab.) was examined. 2. For p-nitrophenylacetate, the Km = 1.24 x 10(-4) M and Vmax = 2.29 x 10(-9) mol/s per mg protein. 3. Regarding four organophosphate insecticides, the mechanism of inhibition in all cases was mixed (competitive and uncompetitive) and, based on inhibition constants, the order of toxicity was naled greater than paraoxon greater than trichlorfon greater than oxydemeton methyl. 4. Comparisons are made to the honey bee, Apis mellifera.

Interaction with host cell receptors initiates internalization of Kaposi's sarcoma-associated herpesvirus (KSHV) particles. Fusion of viral and host cell membranes, which is followed by release of the viral capsid into the cytoplasm, is executed by the core fusion machinery composed of glycoproteins H (gH), L (gL), and B (gB), that is common to all herpesviruses. KSHV infection has been shown to be sensitive to inhibitors of vacuolar acidification, suggestive of low pH as a fusion trigger.

Colorectal cancer (CRC) constitutes the second leading cause of cancer-related death worldwide and advanced CRCs are resistant to targeted therapies, chemotherapies and immunotherapies. p38α (Mapk14) has been suggested as a therapeutic target in CRC; however, available p38α inhibitors only allow for insufficient target inhibition. Here we describe a unique class of p38α inhibitors with ultralong target residence times (designated ULTR-p38i) that robustly inhibit p38α downstream signaling and induce distinct biological phenotypes.

Respiratory syncytial virus (RSV) infection is associated with increased rates of severe disease, hospitalization, and death in elderly individuals. Clearance of RSV is frequently delayed within this demographic, contributing to the more severe disease course. Geriatric cotton rats mimic this prolonged clearance kinetic and serve as a useful animal model for studying age-associated immunological deficits during RSV infection.
