Interferons (IFNs) have antiviral, antimitogenic, and immunostimulatory effects and are often used in the treatment of viral hepatitis and some neoplasms. Combination pegylated IFN-alpha and ribavirin therapy is currently recommended for the treatment of hepatitis C. Triple therapy, with the addition of a protease inhibitor, such as telaprevir or boceprevir, has recently become a mainstay of therapy for certain genotypes. There have also been reports outlining side effects associated with conventional IFN therapy and its immunostimulatory effects, which may cause autoimmune phenomena, including but not limited to Guillain-Barre syndrome, polymyositis, acute and chronic demyelinating polyneuropathy, and myasthenia gravis. Although a number of cases of interferon-induced myasthenia gravis have been reported, we present a case of interferon-induced myasthenia crisis that developed soon after retreatment of hepatitis C with combination interferon, ribavirin, and telaprevir.

Chinese yam polysaccharide (CYP) is an effective immunostimulant, however, its efficacy in grass carp, an important commercial fish species in Asia, remains untested. Here, our study evaluated the immunostimulatory effects of CYP on IgM B cells in vitro and on humoral immunity and immune defense against Aeromonas hydrophila infection in vivo. In vitro stimulation experiments showed that CYP could induce the secretion of IgM antibodies, because it could stimulate the proliferation and differentiation of head kidney IgM B cells.

Background/objectives: Effectively targeting treatment-resistant tumor cells, particularly cancer stem cells (CSCs) involved in tumor recurrence, remains a major challenge in immunotherapy. This study examines the potential of combining mechanical high-intensity focused ultrasound (M-HIFU) with dendritic cell (DC) vaccines to enhance immune responses against OLFM4-expressing tumors, a CSC marker linked to immune evasion and tumor growth.
Methods: M-HIFU was applied to induce immunogenic cell death by mechanically disrupting tumor cells, releasing tumor-associated antigens and creating an immunostimulatory environment.

Synthetic cytosine-phosphate-guanine oligodeoxynucleotides (CpG ODNs) are promising candidates for vaccine adjuvants, because they activate immune responses through the Toll-like receptor 9 (TLR9) pathway. However, unmodified CpG ODNs are quickly degraded by serum nucleases, and their negative charge hinders cellular uptake, limiting their clinical application. Our group previously reported that guanine-quadruplex (G4)-forming CpG ODNs exhibit enhanced stability and cellular uptake.

Bovine alphaherpesvirus-1 (BoAHV-1) causes several symptoms in cattle, leading to significant costs for the livestock industry. In this study, we used a plasmid encoding a secreted form of BoAHV-1 glycoprotein D (pCIgD) as a DNA vaccine. To enhance the potency of the pCIgD vaccine, we used Montanide™ GEL01 PR (GEL01) and introduced Galectin-8 (Gal-8), a lectin considered a novel adjuvant due to its immunostimulatory effects, into the formulation.
