Published: July 2013

A new tetracyclic triterpenoid compound named as mastichinoic acid A (1) was isolated from Mastich. The structure was established as 3,7-dioxo-11β-hydroxy-5α-tirucalla-8,24(Z)-dien-26-oic acid by various spectroscopic techniques including MS, 1D, and 2D NMR and HRTOFMS.

Article Synopsis
  • OSCs (Oxidosqualene cyclases) are important enzymes that change linear triterpenes into tetracyclic compounds, which then serve as precursors for bioactive metabolites.
  • Two OSC genes associated with different synthases, parkeol and lanostadienol, were identified in sea cucumbers from the Sclerodactylidae family to expand knowledge on OSC diversity.
  • Analysis revealed that the two identified OSC genes, OSC1 and OSC2, share a high degree of sequence similarity and suggest distinct evolutionary paths from OSC genes found in the Stichopodidae family.

Typical dammarane-type ginsenosides are well-known tetracyclic triterpenoids with significant pharmacological effects including antitumor, cardiovascular protection, and neuroprotection. Polyene-type ginsenosides exhibit stronger biological activities than common ginsenosides; however, their contents are low, and most are converted from ginsenosides through a series of processing steps, resulting in higher preparation costs. In this study, a dammaradienol synthase, AarOSC20433, was identified for the first time from H.

As one of the metabolites of ginseng, 20(S)-protopanaxadiol (PPD) is a compound with dammarane-type tetracyclic triterpene, which performs a wide range of anticancer activities. In this study, PPD was used as a lead. A series of compounds were synthesized respectively with 11 amino acids through esterification and were evaluated for their cytotoxicity against several cancer cell lines.
