Methemoglobin concentration is an important pathophysiological biomarker, reflecting the oxygen-carrying and oxygen-releasing capabilities of hemoglobin (Hb). Raman spectroscopy is used to develop a novel technique for determining the methemoglobin concentration. Raman activity combined with two-dimensional correlation analysis is an attractive method for investigating Hb oxidation, exhibiting several relevant peaks in the range of 1200-1650 cm(-1). Methemoglobin concentration is estimated by measuring the intensity of Raman peaks in the ranges of 1210-1230 cm(-1) and 1340-1380 cm(-1) with 785-nm excitation. The correlation between Raman-based methemoglobin concentration estimations and the methemoglobin concentration measured using spectrophotometry was highly significant. These results suggest the potential of Raman spectroscopy as a new quantitative approach to determine the methemoglobin concentration.

Background: PAPP is widely used in Australia as a potent vertebrate bait, with potential for off-target ingestion and poisoning in domestic dogs. Whilst toxicosis and resulting methaemoglobinaemia is anecdotally known to occur, this is the first description in the literature. Venous blood is considered an acceptable alternative to arterial blood for assessment of metabolic acid-base disorders. Also, venous sampling using lithium-heparin (Li-Hep) tubes is advantageous to arterial sampling using PICO syringes, the risk of complications being lower. In a previously reported equilibrium study of the reaction of 5,5'-dithiobis(2-nitrobenzoate), DTNB, with various carbonmonoxyhemoglobins over the pH range 5.6 to 9, we obtained contradictory results on the influence of the allosteric effector inositol hexakisphosphate (inositol-P) on the DTNB reaction. For this reason, we replaced the carbonmonoxyhemoglobins with oxyhemoglobins and investigated the effect of inositol-P on the equilibrium and of their reactions with DTNB over the same pH range.

Objectives: Hemolysis is a contributor to CS-AKI. Biochemistry analyzers provide a hemolysis index to quantify in vitro hemolysis, a condition that can, for example, affect the accuracy of potassium concentration measurements.
