A series of fourteen dispiropyrrolidines were synthesized using [3+2]-cycloaddition reactions and were screened for their antimycobacterial activity against Mycobacterium tuberculosis H(37)Rv in HTS (High Throughput Screen). Most of the compounds showed moderate to good activity with MIC of less than 20 μM. Compound 4'-(4-bromophenyl)-1'-methyldispiro[acenaphthylene-1,2'-pyrrolidine-3',2″-indane]-2,1″(1H)-dione (4c) was found to be the most active with MIC of 12.50 μM.

A synthesis of a small library of fluorescent 1,4-dihydropyridine nucleoside analogues has been successfully carried out under solvent-free conditions a one-pot three-component Hantzsch condensation reaction. The process involved a Ba(NO) catalyzed solvent-free reaction between 3',5'-di--acetyl-5-formyl-2'-deoxyuridine, differently substituted β-keto ester and ammonium acetate under microwave irradiation. This facile methodology yielded the desired products with very high yields (86-96%) under solvent-free reaction conditions in a short reaction time, which was followed by a simple workup.

An efficient and facile method has been developed for the construction of novel P-S-C and P-Se-C bonds by facilitating the three-component cross-coupling reaction of P-H bonds with elemental sulfur/selenium and vinylsulfonium salts, utilizing sodium bicarbonate as a base. This approach eliminates the need for the use of toxic and odorous active sulfur/selenium reagents and noble metals, thereby offering a new pathway for synthesizing -phosphinothioates and -phosphinoselenoates via the organic conversion of inorganic sources. The reaction has showcased remarkable versatility in terms of substrate applicability, particularly for organophosphorus compounds containing P-H bonds and vinylsulfonium salt derivatives.

Robust phosphine-based covalent-organic framework palladium catalysts for the highly efficient carbonylation coupling reaction.

Chem Commun (Camb)

December 2024

College of Chemistry and Chemical Engineering, Hunan Provincial Key Laboratory of Micro and Nano Material Interface Science, Central South University, Changsha 410083, Hunan, P. R. China.

The first vinyl-linked PPh-based covalent-organic frameworks (Pd@TMBen-PPh and Pd@TMBen-4F-PPh) were facilely constructed direct three-component Knoevenagel condensation, which could serve as outstanding heterogeneous catalysts for the Pd-catalyzed carbonylation coupling reaction owing to the synergistic effect of the flexible PPh-ligated Pd complexes and the rigid framework.

The DBU-promoted three-component coupling reaction of H-phosphonates, [60]fullerene, and alkyl bromides has been developed as a facile and efficient protocol for the one-pot synthesis of various 1,4-(phosphonyl)(organo)[60]fullerenes. This method exhibits high regioselectivity at the 1,4-position on the [60]fullerene core and features a metal-free nature, easy operation, low cost, and a wide range of substrates.

Center for Metareceptome Research, Graduate School of Pharmaceutical Sciences, Chung-Ang University, 84 Heukseok-ro, Dongjak, Seoul 06974, Republic of Korea.

A visible-light-induced three-component alkoxyalkylation of alkenes has been developed under the photocatalysis of -Ir(ppy). The alkene substrate scope included aryl and aliphatic alkenes as well as electron-rich and electron-deficient alkenes, allowing the facile coupling with a diverse array of α-halocarbonyl compounds. The redox potential-guided orchestration of radical processes with precision allows rapid access to highly functionalized products that are useful building blocks in organic synthesis.
