Hepatic organoid cultures are a powerful model to study liver development and diseases in vitro. However, hepatocyte-like cells differentiated from these organoids remain immature compared to primary human hepatocytes (PHHs), which are the benchmark in the field. Here, we applied integrative single-cell transcriptome and chromatin accessibility analysis to reveal gene regulatory mechanisms underlying these differences.

Lipases such as patatin-like phospholipase domain-containing protein 3 (PNPLA3) exist in multiple tissue types. In subcutaneous adipose tissue, PNPLA3 was not altered during the periparturient period. Conversely, strong associations between liver PNPLA3 and liver triglyceride content peripartum were identified and confirmed to be causative using knockdown approaches in a primary bovine hepatocyte model.

Reliable models of the blood-brain barrier (BBB), wherein brain microvascular endothelial cells (BMECs) play a key role in maintenance of barrier function, are essential tools for developing therapeutics and disease modeling. Recent studies explored generating BMEC-like cells from human pluripotent stem cells (hPSCs) by mimicking brain-microenvironment signals or genetic reprogramming. However, due to the lack of comprehensive transcriptional studies, the exact cellular identity of most of these cells remains poorly defined.

Background/objectives: Hyperlipidemia is a serious risk factor for cardiovascular diseases and liver steatosis. In this work, we explored the effect of an herbal formula (CBF) containing immature pods and extracts on lipid metabolism disorders and lipoprotein-rich plasma (LRP) oxidation in mice.
Methods: The phenolic composition was determined using HPLC-DAD analysis.
