The biochemical roots of the RNA world: from zymonucleic acid to ribozymes.

The history of the ideas that led to the RNA World hypothesis is reviewed. As the understanding of the properties of RNA molecules progressed, the evolutionary interpretation of their genetic properties and widespread distribution in intracellular environments, as well as the catalytic properties of nucleotide coenzymes and the participation of RNA monomers in metabolic pathways, led to several independent proposals of protein-free primordial life forms. Current ideas on the RNA World are part of a long and storied scientific perspective in which these different hypotheses were developed. However, the lack of continuity between them may be explained in part by the absence of an evolutionary framework that characterized the early development of molecular biology, as well as by the demise of certain areas of research like coenzyme chemistry.

Simultaneous quantification of siRNA antisense and sense strands by hybrid liquid chromatography-mass spectrometry.

Background: Most oligonucleotide bioanalytical assays currently only quantify the pharmacologically-active antisense strand, though there have been recent efforts to simultaneously quantify the sense strand using hybridization ELISA or solid phase extraction LC-MS. Hybrid LC-MS, which offers both high sensitivity and specificity unlike the currently used platforms, has not been applied to quantify both siRNA strands simultaneously.

Materials & Methods: A hybrid LC-MS assay utilizing LNA capture probes was developed and applied to quantify both strands of a 21-mer lipid-conjugated siRNA (SIR-3) using tandem mass spectrometry (MS/MS).

Circular RNAs (circRNAs), as a class of noncoding RNA molecules with a circular structure exhibit high stability and spatiotemporal-specific expression, making them ideal cancer biomarkers for liquid biopsy. Herein, a new photoelectrochemical (PEC) biosensor for a highly sensitive circRNA assay in the whole blood of lung cancer patients was designed based on CRISPR/Cas13a-programmed Cu nanoclusters (Cu NCs) and a -scheme covalent organic framework/silver sulfide (T-COF/AgS) composite. This -scheme T-COF/AgS composite accelerates electron transfer and produces an excellent initial photocurrent.

Brassinosteroids (BRs) are key phytohormones influencing soybean development, yet their role in symbiosis remains unclear. Here, the RNA-Seq was used to identify important gene associated with BRs and symbiotic nitrogen fixation, and the function of candidate gene was verified by transgenic hairy roots. The result shows that the RNA-Seq analysis was conducted in which BR signaling was found to suppress nodule formation and many DEGs enriched in immunity-related pathways.
