Hemicalide is a novel marine metabolite polyketide distinguished by a unique mechanism of action. Because of insufficient quantities of purified material, this natural product has evaded complete stereochemical assignments. Recently, we have determined the relative stereochemistry of the C8-C13 hexad by synthesizing the C1-C13 fragment. Presently, we report the assignment of the C17-C25 δ-lactone fragment. NMR analysis of authentic hemicalide along with a computational conformation study allowed us to reduce the number of putative relative isomers from 16 to 4. Concise syntheses of the four candidate diastereomers were achieved using a common strategy based on a Dias aldehyde allylation reaction, an intramolecular Horner-Wadsworth-Emmons olefination, and a dihydroxylation reaction. Finally, thorough NMR comparisons enabled us to deduce the relative stereochemistry of the C1-C17 fragment with high certainty.

Eighteen polycyclic aromatic hydrocarbons (PAHs), 24 n-alkanes, 7 hopanes, 2 cholestanes, inorganic ions, elements and carbon fractions were analyzed in real-world source samples of PM (fine particulate matter) from traffic emissions (gasoline vehicles-TGV, diesel vehicles-TDV, diesel ship-TDS, and heavy oil ships-THOS), coal combustion (coal-fired industrial boilers-CIB, power plants-CPP, and residential stoves-CRS), industrial process emissions (cement industry-IPCI, and steel industry-IPSI), and dust (soil dust-DSD, road dust-DRD, and construction dust-DCD). High molecular weight (sum of five to seven rings) PAHs accounted for higher fractions for TGV (80%) and THS (61%) than for TDV, TDS and coal combustion sources (31%-47%). Hopane ratios (C29αβ/C30αβ) in coal related sources were mostly higher than 1, whereas that of traffic emissions was lower than 1.

Sleep deprivation has profound influence on several aspects of health and disease. Preliminary studies into the use of ring-closing metathesis (RCM) in a convergent approach for the total synthesis of bryostatins are described. An ester that would have provided an advanced intermediate for a synthesis of a 20-deoxybryostatin by a RCM was prepared from an unsaturated acid and alcohol corresponding to the C1-C16 and C17-C27 fragments. However, studies of the formation of the C16-C17 double-bond by RCM were not successful and complex mixtures of products were obtained.
