Objective: This study aims to explore the potential role of mesenchymal stem cells (MSCs) in the treatment of osteoarthritis (OA), particularly the function of the NOTCH1 signaling pathway in maintaining the stemness of MSCs and in chondrocyte differentiation.
Methods: Utilizing diverse analytical techniques on an osteoarthritis dataset, we unveil distinct gene expression patterns and regulatory relationships, shedding light on potential mechanisms underlying the disease. Techniques used include the culture of MSCs, induction of differentiation into chondrocytes, establishment of stable cell lines, Western Blot, and immunofluorescence.

Objective: Interleukin-17 E (IL-17E) is a pro-inflammatory cytokine that participates in the inflammatory response and tumorigenesis. However, the function of IL-17E in non-small cell lung cancer (NSCLC) remains largely unknown.
Methods: The clinical value of IL-17E was determined by immunohistochemistry (IHC) in 75 cases of NSCLC tissues.

This is a maximal intensity projection of CA1 pyramidal cell transfected with plasmid with the reporter GFP using single cell electroporation technique. In this particular case the organotypic slices were prepared from p5-7 pups in a tissue chopper (McIlwain). And maintained in MEM bases media with added glutamax with a change in 2 alternative dyas at 37°C and 5% CO for 4 days in-vitro (DIV) before electroporating with a glass pipette of 7-10mΩ resistance by applying 4 square pulses of -ve voltage of -2.

Background: Rodent models have been proved pivotal in Alzheimer's disease (AD) research. Nevertheless, the use of models that only recapitulate one aspect of AD neuropathology, and of early time points that might be excluding important features such as age-dependent inflammation and senescence, could hinder the development of effective AD therapeutics. Several tau immunotherapies are currently undergoing clinical trial.

Background: In Alzheimer's disease (AD), specific brain regions become vulnerable to pathology while others remain resilient. New methods of imaging such as highly multiplexed immunofluorescence (MxIF) provide an abundance of spatial information, while analytical techniques like machine learning (ML) can address questions of cellular contributors to this regional vulnerability.
Method: We performed MxIF staining for 26 markers and compared postmortem human samples from an AD-susceptible brain area, the prefrontal cortex (PFC, Brodmann's areas 9, 10 or 46) to an AD-resilient brain area, the primary visual cortex (V1, area 17).
