The rockfish Sebastes schlegeli skin mucus contains a potent antibacterial protein, SSAP (S. schlegeli antibacterial protein), a novel l-amino acid oxidase with strict substrate specificity that acts against water-borne Gram-negative bacteria. We previously demonstrated that SSAP distributes in the skin and gills. Here we investigated the intra-tissue localization of SSAP in the tissues by in situ hybridization. Skin and gill sections were hybridized with digoxigenin-conjugated SSAP-specific RNA probe. SSAP mRNA-positive cells located near the basal membrane of skin epidermis and the gill epithelium. Furthermore, skin section was analyzed by immunohistochemistry and reacted with anti-SSAP antiserum as a primary antibody. The mucus layer and mucous cells in the skin were immunopositive. Skin and gill extracts produced hydrogen peroxide, responsible for antibacterial activity, in the presence of l-lysine. These results suggested that SSAP functions locally as a humoral defense factor in S. schlegeli skin and gills and prevents pathogenic bacterial invasion.

Determining whether an ectopic depolarization will lead to a self-perpetuating arrhythmia is of critical importance in determining arrhythmia risk, so it is necessary to understand what factors impact substrate vulnerability. This study sought to explore the impact of cell-to-cell heterogeneity in ion channel conductance on substrate vulnerability to arrhythmia by measuring the duration of the vulnerable window in computational models of one-dimensional cables of ventricular cardiomyocytes. Objectives: Cartilage targeting cationic glycoprotein Avidin was PEGylated to synthesize a multi-arm Avidin (mAv) nano-construct with high drug loading content. Here we investigate mAv biodistribution and kinetics over a 7-day period following intra-articular (IA) administration in rat knee joints.
Methods: Labeled mAv was injected into healthy rat knees, and joint tissues (articular cartilage, menisci, ligaments, tendons, fat pad) were harvested following sacrifice at 6 h, 1, 4 and 7 days.

Dehydroepiandrosterone (DHEA), an adrenal androgen precursor, can be metabolized in target tissues into active sex steroids. It has been proposed that DHEA supplementation might result in restoration of physiological local sex steroid levels, but knowledge on the effect of DHEA treatment on local sex steroid levels in multiple tissues is lacking.
