Kinetic stabilization of amyloidogenic immunoglobulin light chains (LCs) through small molecule binding may become the first treatment for the proteinopathy component of light chain amyloidosis (AL). Kinetic stabilizers selectively bind to the native state over the misfolding transition state, slowing denaturation. Prior λ full-length LC dimer (FL LC) kinetic stabilizers exhibited considerable plasma protein binding.

Aim: Oxidative stress, caused by postprandial activities, is a major global health issue causing chronic diseases like diabetes mellitus, cancer, and asthma. Therefore, it was envisaged to design and synthesize a series of substituted 4-hydroxypyridine-2(1 h)-ones in order to develop new molecules that can reduce oxidative stress and modulate α-amylase activity also.

Materials & Methods: An environmentally benign, solvent and catalyst free, natural product inspired synthesis of 4-hydroxypyridin-2(1 h)-one derivatives has been developed.

KOBu-Promoted [4+2] Annulation-Dehydration Cascade Enabling the Construction of Diverse 2-Pyridone-Fused Uracils.

Org Lett

December 2024

Department of Chemistry and Research Institute for Natural Sciences, Hanyang University, Seoul 04763, Korea.

A KOBu-promoted [4+2] annulation-dehydration cascade reaction has been developed, enabling the efficient synthesis of diverse 2-pyridone-fused uracils through a vinylogous enolization strategy involving -quinodimethane (QDM) dienolate intermediates. This method provides a simple yet robust approach for constructing structurally interesting fused -heterocycles that incorporate two privileged scaffolds, both of which are widely recognized in drug discovery. Consequently, these compounds hold significant potential for biological and pharmacological applications.

2-Pyridone-Enhanced Mn-Catalysis for the Synthesis of -Deuterated Aromatic Nitriles.

Org Lett

November 2024

NingboTech-Cuiying Joint Laboratory of Stable Isotope Technology, School of Biological and Chemical Engineering, NingboTech University, Ningbo 315100, People's Republic of China.

We herein present a method for the synthesis of -deuterated aryl nitrile using Mn(CO)Br as catalyst with CHOD as deuteratium source, where the structure of aryl imidates is used for interconversion with a cyanide group. This method features a broad substrate range and excellent functional group tolerance, allowing late modification of various complex molecules with good yields and deuterium incorporation. Mechanistic studies suggest that 2-pyridone is crucial to the success of this chemistry, serving as an endogenous base that enhances the rate of hydrogen isotope exchange.

Singlet oxygen storage and controlled release for improving photodynamic therapy against hypoxic tumor.

Chem Commun (Camb)

November 2024

School of Chemistry and Chemical Engineering, Nantong University, Nantong, Jiangsu, 226019, P. R. China.

Photodynamic therapy (PDT) is considered to be a promising tumor treatment method due to its non-invasiveness and low risk. However, there are two factors that affect the efficacy of this therapy. One is the light source and the other is the tumor hypoxia.
