Comparison of photo- and Cu(I)-catalyzed "click" chemistries for the formation of carbohydrate SPR interfaces.

Understanding interactions of glycans with proteins in key biological events has seen the development of various analytical methods such as microarray techniques. Label-free approaches, such as surface plasmon resonance (SPR) techniques are particularly attractive and we explore here the potential of a novel interface composed of lamellar Ti/Au/silicon dioxide derivatized with sugars to probe lectin-sugar interactions by SPR. Two parallel surface functionalization strategies have been developed: one in which azide-functionalized surfaces are linked via a Cu(I) "click" method to alkynyl-derivatized glycan partners and another wherein perfluorophenyl azide-functionalized surfaces are reacted through a C-H insertion photocoupling reaction with underivatized glycans. The effectiveness of the two interfaces is assessed for their lectin-recognition abilities in an SPR format.

Cellulose tosylate (MCC-Tos) is a key derivative for surface modification and a crucial precursor for cellulose compatibilization in click reactions, enabling its functionalization for advanced applications. Therapeutic proteins are commonly conjugated with polymers to modulate their pharmacokinetics but often lack a description of the polymer-protein interaction. We deployed limited proteolysis mass spectrometry (LiP-MS) to reveal the interaction of polyethylene glycol (PEG) and PEG alternative polymers with interferon-α2a (IFN). Target conjugates were digested with the specific protease trypsin and a "heavy" N-IFN wild type (IFN-WT) for time-resolved quantification of the cleavage dynamics.

A simple and sensitive method for Cu detection was developed using the Cu-catalyzed alkyne-azide cycloaddition reaction, FeO magnetic nanoparticles (MNPs) as the reaction platform, and a portable blood glucose meter (PGM) as the detection method. Gold nanoparticles (AuNPs) were labeled with glucose oxidase (GOx) and alkyne-functionalized, terminally thiolated ssDNA (C2). In the presence of Cu and ascorbate, the functionalized AuNPs were captured onto MNPs modified with azide-functionalized ssDNA (C1) via the Cu-catalyzed alkyne-azide cycloaddition reaction.
