Interpreting the biological implications of high-throughput experiments such as gene-expression studies, genome-wide association studies and large-scale sequencing studies is not trivial. Gene-set and pathway analyses are useful tools to support the interpretation of such experiments, but rely on curated pathways or gene sets. The recent development of de novo pathway discovery methods aims to overcome this limitation. This article provides an overview of the methods currently available and reviews the advantages and challenges of this approach. In detail, it highlights the particular issues of de novo pathway discovery based on genome-wide association studies data, for which multiple different strategies have been proposed.

Aim: The goal of this study was to determine the role of histone deacetylase 9 (HDAC9) in the development of diet-induced metabolic dysfunction-associated steatohepatitis (MASH) and white adipose tissue (WAT) dysfunctions.
Methods: We fed male and female mice with global Hdac9 knockout (KO) and their wild-type (WT) littermates an obesogenic high-fat/high-sucrose/high-cholesterol (35%/34%/2%, w/w) diet for 20 weeks.
Results: Hdac9 deletion markedly inhibited body weight gain and liver steatosis with lower liver weight and triglyceride content than WT in male mice but not females.

The capacity for a non-native species to become invasive largely hinges on existing dispersal capacity or adaptation of dispersal in new environments. Here we provide early evidence that invasive Northern Pike (Esox lucius), a Holarctic freshwater top predator, illegally introduced in the late 1950s into Southcentral Alaska, are now dispersing through estuarine corridors. This finding represents the first known documentation of estuary use and dispersal by Northern Pike in North America, exacerbating conservation concerns for already depressed populations of culturally and economically important species such as salmonids.
