The hydrothiolation of unsaturated carbon-carbon bonds is a practical and atom-economical approach for the incorporation of sulfur into organic frameworks. In recent years, we have witnessed the development of a range of transition-metal-based catalytic systems for the control of the regio- and stereoselectivity. In this Minireview we highlight the mechanistic background behind this transformation so as to help the design of more specific and active organometallic hydrothiolation catalysts.

For the first time, ball milling has been employed in the solvent-free synthesis of sulfur-functionalized materials from thiols and α,β-unsaturated ketones and esters, using potassium carbonate as a transition metal-free catalyst. This environmentally friendly protocol makes use of easily accessible reagents to prepare thirty β-sulfenylated carbonyl compounds with yields exceeding 91 % under ambient air and solvent-free conditions. Additionally, this innovative synthetic strategy enables the modification of chalcones, compounds with significant medicinal and synthetic potential.

The addition of heteroatom compounds to alkynes and alkenes is an atom-efficient method of carbon-heteroatom bond formation and is widely used as a fundamental synthetic method for the construction of functional molecules. Nevertheless, examples of transition-metal-catalyzed addition reactions of group 16 heteroatom compounds to carbon-carbon unsaturated bonds have been limited due to the widespread belief that organic sulfur and selenium compounds are representative catalyst poisons. In recent decades, however, several seminal catalytic reactions of sulfur compounds have been developed, providing important insights into catalysis and poisons.

Synthesis of 4-thiol-furanosidic uronate hydrothiolation reaction.

Uronic acids are not only important building blocks of polysaccharides and oligosaccharides but also are widely used in the food and pharmaceutical industries. Inspired by the structure of natural products, here, we disclosed base-mediated and radical-mediated hydrothiolation reactions for the preparation of thiol-contained uronates. In comparison with base-mediated reaction, radical-mediated hydrothiolation is inefficient due to the electron-withdrawing group on the ethylene group; nevertheless, the adduct had excellent stereoselectivity at both C-4 and C-5 positions.

Sulfur centered radicals are widely employed in chemical synthesis, in particular for alkene and alkyne hydrothiolation towards thioether bioconjugates. The steadfast radical chain process that enables efficient hydrothiolation has been explored in the context of cascade reactions to furnish complex molecular architectures. The use of thiyl radicals offers a much cheaper and less toxic alternative to the archetypal organotin-based radical methods.
