Protoplasts were prepared from tubers of Helianthus tuberosus in the developing and in the resting stage of development. Vacuoles were isolated from the protoplasts and purified by sedimentation through a density gradient of glycine betaine. All the fructan (inulin) with a DP ⩾ 3 (i.e. the trisaccharide isokestose and larger polymers of fructose) was found to be located exclusively in the vacuoles whilst sucrose, glucose, and fructose were located only partially there. The vacuoles were also found to be the sole cell compartments containing fructan synthesizing enzyme activities (sucrose-sucrose-fructosyl-transferase and fructan-fructan-fructosyl-transferase) and fructan degrading enzyme activity (fructan exohydrolase), depending on the stage of development of the tubers. The vacuole is therefore proposed to be the cell organelle for fructan storage and metabolism.

In cervical cancer screening, cytology is used as a triage test to refer high-risk human papillomavirus (HR-HPV)-positive women for colposcopy, but its accuracy is inadequate. The present study aimed to demonstrate that the presence of atypical cells with large vacuoles in the cytoplasm of parabasal cells, referred to as vacuolated parabasal cells (VPCs), which are observed in the Pap smears of HPV-positive women, is associated with specific HPV genotypes. Among 2175 patients, 310 with a single HR-HPV infection and cytological diagnosis of high-grade squamous intraepithelial lesions (HSIL) or atypical squamous cells not excluding HSIL (ASC-H) were included, of which 86 were infected with HPV16.

During routine sampling of northern pike, a male with circular blue-metallic granular spots mainly located on the head and back was identified. Histological investigations presented multifocally thickened epidermis rich in basophilic large structures with a granulated rim and a dense, non-granulated center.
