Unveiling the Emerging Role of Klotho: A Comprehensive Narrative Review of an Anti-aging Factor in Human Fertility.

Curr Protein Pept Sci

September 2024

Department of Obstetrics and Gynecology, All India Institute of Medical Sciences, Bibinagar-508126, Hyderabad, Telangana, India.

Klotho, an anti-aging protein, plays a vital role in diverse biological functions, such as regulating calcium and vitamin D levels, preventing chronic fibrosis, acting as an antioxidant and anti-inflammatory agent, safeguarding against cardiovascular and neurodegenerative conditions, as well as exerting anti-apoptotic, anti-senescence effects. Additionally, it contributes to metabolic processes associated with diabetes and exhibits anti-cancer properties. This protein is commonly expressed in organs, such as kidneys, brain, pancreas, parathyroid glands, ovaries, and testes.

Importance: Delayed autotransplantation of cryopreserved parathyroid tissue (DACP) is the only surgical treatment for permanent postoperative hypoparathyroidism. Studies suggest that only a small minority of cryopreserved samples are ultimately autotransplanted with highly variable outcomes. For these reasons, many have questioned the economic utility of the process, although, to the authors' knowledge, this has never been formally studied.

Metabolism of parathyroid organoids.

Front Endocrinol (Lausanne)

July 2023

Division of Surgical Oncology & Endocrine Surgery, Department of Surgery, Vanderbilt University Medical Center, Nashville, TN, United States.

Introduction: We successfully developed a broad spectrum of patient-derived endocrine organoids (PDO) from benign and malignant neoplasms of thyroid, parathyroid, and adrenal glands. In this study, we employed functionally intact parathyroid PDOs from benign parathyroid tissues to study primary hyperparathyroidism (PHPT), a common endocrine metabolic disease. As proof of concept, we examined the utility of parathyroid PDOs for bioenergetic and metabolic screening and assessed whether parathyroid PDO metabolism recapitulated matched PHPT tissues.

Multiple endocrine neoplasia type 2A (MEN2A) is a rare hereditary condition characterized by medullary thyroid cancer, pheochromocytoma, and primary hyperparathyroidism. The current standard of treatment of hyperparathyroidism involves surgical removal of visibly enlarged glands, and auto-transplantation of remnant tissue is often considered to minimize the risk of iatrogenic post-surgical hypocalcemia if multiple glands are enlarged. Rarely, hyperparathyroidism may recur due to hyperplasia or adenoma formation in the auto-transplanted tissue.
