This article presents models to predict mutagenicity, carcinogenicity, developmental toxicity, and skin sensitisation for a set of 27 conazoles. The predictions were performed with the program package CAESAR, which is available on the Internet. The CAESAR programs were developed to support the European Community Regulation on chemicals and their safe use (REACH) and follow the OECD principles for (Q)SAR models used for regulatory purposes. The programs provide a number of information, including a binary classification of a compound as toxic or non-toxic and information on similar compounds from the model's training sets (similarity sets). In this study we analysed conazole sets using principal component analysis (PCA). The predictions were compared to the currently valid classification of these substances in the European Union (EU) or to the classification proposed at expert meetings of the Pesticide Risk Assessment and Peer Review (PRAPeR) group. The predicted classification for mutagenicity was in good agreement with regulatory classification, the predictions for carcinogenicity and developmental toxicity showed some discrepancy in particular cases, while the predictions for skin sensitisation showed even greater discrepancy.

N-Nitrosodimethylamine (NDMA) is a known water disinfection byproduct (DBP) characterized as a potent hepatotoxin, promutagen, and probable human carcinogen; this is because of the metabolites associated with its biotransformation. The metabolism of NDMA produces formaldehyde, another alkylating agent and DBP. Both compounds are generated from natural and anthropogenic sources, but the safety restrictions applied to NDMA do not extend to the uses of formaldehyde.

In the shipbuilding industry, during the painting process, workers are exposed to various substances in paint, including organic solvents that can adversely affect their health. Most workplace exposures to organic solvents involve mixtures of organic compounds. Therefore, in this study, the hazard quotient (HQ) and hazard index (HI) were derived using data from the Workplace Environmental Monitoring Program in Korea for six organic solvents (xylene, n-butanol, ethylbenzene, isobutyl alcohol, toluene, and methylisobutyl ketone [MIBK]) commonly used in the steel shipbuilding industry.

Marketed endothelin receptor antagonists (ERAs) have been associated with testicular tubular atrophy and decreases in male animal fertility in chronic toxicity studies in rats and dogs. Consistent with these findings, reduced sperm count has been observed in the clinical setting and is considered a potential class risk with chronic administration of ERAs. In contrast, no such effects on male animal fertility are noted with angiotensin II type 1 receptor blocker (ARB) treatment.
