The genome of vertebrates contains endogenous retroviruses (ERVs) that are largely nonfunctional relicts of ancestral germline infection by exogenous retroviruses. However, in some mouse strains ERVs are actively involved in disease. Here we report that nucleic acid-recognizing Toll-like receptors 3, 7, and 9 (TLR 3, TLR7, and TLR9) are essential for the control of ERVs. Loss of TLR7 function caused spontaneous retroviral viremia that coincided with the absence of ERV-specific antibodies. Importantly, additional TLR3 and TLR9 deficiency led to acute T cell lymphoblastic leukemia, underscoring a prominent role for TLR3 and TLR9 in surveillance of ERV-induced tumors. Experimental ERV infection induced a TLR3-, TLR7-, and TLR9-dependent group of "acute-phase" genes previously described in HIV and SIV infections. Our study suggests that in addition to their role in innate immunity against exogenous pathogens, nucleic acid-recognizing TLRs contribute to the immune control of activated ERVs and ERV-induced tumors.

Background: Although most patients with diffuse large B-cell lymphoma (DLBCL) achieve complete remission after first-line rituximab-containing immunochemotherapy, up to 40% of patients relapse and require salvage therapy. Among those patients, a substantial proportion remain refractory to salvage therapy due to insufficient efficacy or intolerance of toxicities. A hypomethylating agent, 5-azacytidine, showed a chemosensitizing effect when primed before chemotherapy in lymphoma cell lines and newly diagnosed DLBCL patients.

To investigate the pathogenic risk of endogenous retroviruses (ERVs) infection in immunodeficient hosts, the ERV of N-type ecotropic murine leukemia virus (MuLV) isolated from SL mice, a kind of mice containing considerable infectious ERV particles determined with SC-XC test and developing leukemia spontaneously with average of high frequency of 30% and incubation period of 315days, was inoculated intraperitoneally into newborn CBA nude mice. The distinct marker of splenomegaly for leukemia was observed in 33% of homozygous (nu/nu) and 17% of heterozygous (nu/+) of CBA nude mice with average incubation period of 310days and 432days post-inoculation, respectively. Furthermore, the ERV induced leukemia in both the SL mice and CBA nude mice was identified to be B lymphatic, transplantable and with rearrangement of the Evi-1 locus.
