The percentage of metmyoglobin (%metMb) in aqueous meat extracts of bigeye and bluefin tuna and beef samples were estimated using previously reported equations derived from the absorption spectra of horse Mb. The results demonstrate that in an aqueous extract, the difference in %metMb estimated by the different equations was negligible for beef samples. Conversely, in an aqueous tuna extract, different %metMb values were obtained with the different equations. The discrepancy in the tuna sample results might be due to differences in absorption spectra for horse and tuna Mb. Therefore, a new set of equations derived from the absorption spectra of bigeye tuna Mb, reported by Matsuura and Hashimoto (1955), was established. The accuracy of the proposed equations was compared with the cyanmetmyoglobin (cyanmetMb) method. The results show that the total Mb concentrations estimated by our proposed equations were in good agreement with the results obtained by the conventional cyanmetMb method (R(2)=0.984). Therefore, the new set of proposed equations is valid for the spectrophotometric determination of the relative proportions of Mb derivatives and total Mb concentration in aqueous tuna meat extracts.

A novel approach for reducing mercury content in fish meat during post-packaging storage is developed to extend the margin of their safe consumption. It involves employing a single-component aqueous medium containing cysteine, as the active agent responsible for displacing mercury from fish proteins and its stabilization in the medium without the need for pH adjustments. The mercury removal efficiency depends on the cysteine concentration and its ratio to fish muscle.

Two potent antioxidants, selenoneine and ergothioneine, have been determined simultaneously for the first time in the muscle of highly consumed fish and in fish-derived products. The extraction methods were optimized, and their subsequent analysis was performed by HPLC-ESI-MS/MS. Best extraction efficiency (106 ± 3 and 104 ± 5%) was attained by performing a simple aqueous extraction containing 100 mmol L ammonium acetate or ammonium formate as extractants.

The electrical impedance of dilute aqueous solutions containing extracts from five brands of canned tuna is analyzed using impedance spectroscopy in order to analyze their salt content and detect the potential presence of other salts beyond the well-stated NaCl. A complex electrical impedance is modeled with an equivalent electrical circuit, demonstrating good agreement with experimental data. This circuit accounts for the contribution of ions in the bulk solution, as well as those contributing to electrode polarization.
