The multilineage potentiality of cord blood stem cells has been experimentally proven in a number of cell based therapies. Umbilical cord blood (UCB) derived mesenchymal stem cells (MSCs), on prolonged exposure with Lascorbic acid have been successfully differentiated in to osteoblasts (bone forming cells) without altering the phenotype of the cells. In this case study, the role of L-ascorbic acid on collagen biosynthesis and mineral deposition in MSCs has been assessed, which are ultimately matured in to an insoluble extra cellular matrix (ECM), giving mechanical strength to the bone cells. Moreover, up to specific concentration of L-ascorbic acid (250μM), proliferation as well as differentiation potential of the cells remains unaltered. Further increase in concentrations of L-ascorbic acid (500 μM) reduced the cell proliferation and subsequently leads to morphological changes in the cultures. This may be due to an immature antioxidant defense system, which can be overcome by treating the cell cultures with antioxidants. Our final results conclude that Lascorbic acid has positive effect on the ostogenic differentiation of cord blood stem cells, and the concentration of ascorbic acid is vital in cell proliferation and differentiation.

The traditional approach to resuscitating injured women of childbearing potential (WCBP) with an unknown RhD type is to transfuse RhD-negative blood products. This is to prevent alloimmunisation to the RhD antigen and ultimately prevent haemolytic disease of the fetus and newborn (HDFN) in future pregnancies should she survive. RhD-negative blood products are scarce in both military and civilian blood stocks.

Although the corticosteroid betamethasone is routinely administered to accelerate lung and cardiovascular maturation in the preterm fetus prior to birth, and use of delayed cord clamping (DCC) is recommended at birth by professional bodies, it is unknown whether antenatal betamethasone alters perinatal pulmonary or systemic arterial blood flow accompaniments of DCC. To address this issue, preterm fetal lambs [gestation 127 (1) days, term = 147 days] with (n = 10) or without (n = 10) antenatal betamethasone treatment were acutely instrumented under general anaesthesia with flow probes to obtain left (LV) and right ventricular (RV) outputs, major central arterial blood flows and shunt flow across both the ductus arteriosus and foramen ovale (FO). After delivery, lambs underwent initial ventilation for 2 min prior to DCC.

Background: Ferroptosis and immune responses are critical pathological events in spinal cord injury (SCI), whereas relative molecular and cellular mechanisms remain unclear.
Methods: Micro-array datasets (GSE45006, GSE69334), RNA sequencing (RNA-seq) dataset (GSE151371), spatial transcriptome datasets (GSE214349, GSE184369), and single cell RNA sequencing (scRNA-seq) datasets (GSE162610, GSE226286) were available from the Gene Expression Omnibus (GEO) database. Through weighted gene co-expression network analysis and differential expression analysis in GSE45006, we identified differentially expressed time- and immune-related genes (DETIRGs) associated with chronic SCI and differentially expressed ferroptosis- and immune-related genes (DEFIRGs), which were validated in GSE151371.
