Introduction: A robust, versatile and compact microreactor has been designed, fabricated and tested for the labeling of bifunctional chelate conjugated biomolecules (BFC-BM) with PET radiometals.
Methods: The developed microreactor was used to radiolabel a chelate, either 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) or 1,4,7-triazacyclononane-1,4,7-triacetic acid (NOTA) that had been conjugated to cyclo(Arg-Gly-Asp-DPhe-Lys) peptide, with both ⁶⁴Cu and ⁶⁸Ga respectively. The microreactor radiolabeling conditions were optimized by varying temperature, concentration and residence time.
Results: Direct comparisons between the microreactor approach and conventional methods showed improved labeling yields and increased reproducibility with the microreactor under identical labeling conditions, due to enhanced mass and heat transfer at the microscale. More importantly, over 90% radiolabeling yields (incorporation of radiometal) were achieved with a 1:1 stoichiometry of bifunctional chelate biomolecule conjugate (BFC-BM) to radiometal in the microreactor, which potentially obviates extensive chromatographic purification that is typically required to remove the large excess of unlabeled biomolecule in radioligands prepared using conventional methods. Moreover, higher yields for radiolabeling of DOTA-functionalized BSA protein (Bovine Serum Albumin) were observed with ⁶⁴Cu/⁶⁸Ga using the microreactor, which demonstrates the ability to label both small and large molecules.
Conclusions: A robust, reliable, compact microreactor capable of chelating radiometals with common chelates has been developed and validated. Based on our radiolabeling results, the reported microfluidic approach overall outperforms conventional radiosynthetic methods, and is a promising technology for the radiometal labeling of commonly utilized BFC-BM in aqueous solutions.

Currently, metal-organic frameworks (MOFs) with tunability and covalent organic frameworks (COFs) with high stability are promising nanomaterials for electrochemiluminescence (ECL), while Ru-based metal covalent organic frameworks (Ru-MCOFs) have rarely been reported. Herein, an ECL immunosensor based on a strong ECL-emitting Ru-MCOF was proposed for the sensitive detection of the cardiac troponin-I (cTnI). Imine-linked Ru-MCOF was prepared as an ECL emitter via solvothermal method using tris(4,4' -diamino-2,2' -bipyridine)ruthenium(II) (Ru(dbpy)32+) as a precursor.

The selective decarboxylative fluorination of 3-oxo-3-phenylpropionic acid is used as a benchmark reaction to optimize it under biocompatible conditions in batch and to transfer it to continuous flow mode. The reaction conditions are varied with respect to temperature, fluorinating reagents, inorganic base additives, and pH, as these parameters have been identified as having a significant impact on the process. The formation of the products and any by-products is analyzed using gas chromatography (GC) and 19F nuclear magnetic resonance spectroscopy (NMR).

Advanced porous nanomaterials have recently been the subject of considerable interest due to their high surface areas, tunable pore structures, high porosity, and ease of modification. In the chemiluminescence (CL) domain, the incorporation of additional pores into nanostructures not only enhances the loading capacity for signal amplification but also allows the confinement effect in a nanoscale microreactor and the controlled release of reaction agents. Several TRCEM methods have been developed in the past to obtain key insights into the mechanism of action of molecules and molecular machines on the time scale of tens to hundreds of milliseconds, which is unattainable by the normal blotting method. Here we present our TRCEM setup utilizing a polydimethylsiloxane (PDMS)-based microfluidics chip assembly, comprising three components: a PDMS-based, internally SiO -coated micromixer, a glass-capillary microreactor, and a PDMS-based microsprayer for depositing the reaction product onto the EM grid.

In an effort to balance energy and molecular stability effectively, several energetic compounds (-) based on benzotriazine were designed and synthesized. These structures were comprehensively characterized using NMR, IR, and elemental analysis, with compounds , , and further confirmed by single-crystal X-ray diffraction. Notably, 3-amino-5,7-dinitrobenzo[][1,2,4]triazine 1-oxide (), which features a face-to-face crystal stacking arrangement, exhibits good detonation velocity ( = 8050 m/s), a high thermal decomposition temperature ( = 290 °C), and low sensitivities (impact sensitivity >40 J, friction sensitivity >360 N).
