A novel and efficient synthetic approach to substituted 2-iodo-spiro[indene-1,1'-isobenzofuran]-3'-ones has been developed via an iodine-promoted cascade cyclization of 2-(3-hydroxy-3,3-diarylprop-1-yn-1-yl)benzoates. This sequential cascade process is concisely conducted at room temperature. Subsequent palladium-catalyzed Sonogashira, Suzuki, and Heck reactions of the resulting iodides proceed smoothly in good yields.

We report a metal additive-free iodine-promoted one-step structural reorganization of ynamide-ynes and simultaneous stereoselective 1,2-diiodination of the migrated alkyne to form stereospecific tetrasubstituted alkenyl diiodo-tethered indoles (E-isomer). Molecular iodine is cost effective, user friendly, less toxic, commercially available, and easy to handle. The key features of the reaction include metal-and additive-free environment, selectivity, structural reorganization, mild reaction conditions, simple workup, and gram-scale synthesis.

Herein, we report an efficient synthetic method for constructing the tetracyclic scaffold of sulfur-containing discorhabdin-type alkaloids. The key to success is the BF•EtO-promoted dienone-phenol-type rearrangement/sulfur insertion cascade reaction, which converts the common and readily available -acetal-bridged tetracyclic framework to the labile and synthetically challenging -acetal-bridged tetracyclic framework in a single step. Additionally, the hypervalent iodine promoted intramolecular oxidative dearomatization terminated with amide and indium(III) acetate-facilitated radical cyclization were also essential elements in this study.

An unexpected iodine()-mediated C(sp)-C(sp) bond cleavage of 3-(methylamino)-2-(2-substitutedbenzoyl)acrylates for efficient synthesis of privileged scaffold 4-quinolones was described. Notably, a wide range of alkyl groups ( methyl, -butyl or alkyl chain) can be conveniently cleaved in this system. The detailed mechanism studies revealed that the transformation proceeded through cascade ipso-cyclization and 1,2-carbonyl migration, the smaller bond energy determined C-C bond cleavage rather than C-H bond cleavage, an enamine radical intermediate.

An iodine-promoted one-pot cascade oxidative annulation reaction has been developed for the synthesis of chromone-fused-pyrrolo[2,1-]isoquinolines and indolizino[8,7-]indoles from -acetylphenoxyacrylates, tetrahydroisoquinolines, and noreleagnines. This process underwent a logical approach to both chromone-fused-pyrrolo[2,1-]isoquinolines and chromone-fused-indolizino[8,7-]indoles isolamellarin derivatives. Manipulations of l-menthol and dl-α-tocopherol demonstrate the applications of this strategy.

An iodine promoted cascade cycloisomerization of 1-en-6,11-diynes is presented for the easy preparation of tetrahydrobenzo[f]isoquinolines. This developed reaction system is identified as having good functional-group applicability and can be scaled up to gram quantities. In this transformation, two new cyclic frameworks and one carbonyl group are formed with four new bonds constructed.
