In all tissues the balance of cell proliferation and differentiation needs to be tuned to match the varying requirements of embryonic development and adult life. This is well illustrated by the interfollicular epidermis (IFE), which undergoes expansion and remodeling in utero, significant post natal growth and is then maintained in homeostasis. In addition to sustaining a high daily turnover of cells, the epidermis is able to re-populate areas of tissue damage due to common environmental stresses such as wounding. Here recent insights into proliferating cell behavior in IFE and how this changes through development and into adulthood are discussed.

Despite the pivotal role of cytotoxic T lymphocytes (CTLs) in anti-tumor immunity, a substantial proportion of CTL-rich hepatocellular carcinoma (HCC) patients experience early relapse or immunotherapy resistance. However, spatial immune variations impacting the heterogeneous clinical outcomes of CTL-rich HCCs remain poorly understood. Lineage tracing based on modern live imaging approaches enables to visualize, reconstruct, and analyze the developmental history, fate, and dynamic behaviors of cells in vivo in a direct, comprehensive, and quantitative manner. Light-sheet fluorescence microscopy (LSFM) has greatly boosted lineage tracing efforts, because fluorescently labeled specimens can be imaged in their entirety, over long periods of time, with high spatiotemporal resolution and minimal photodamage. In addition, an increasing arsenal of commercial and open-source software solutions for cell and nuclei segmentation and tracking can be employed to convert data from pixel-based to object-based representations, and to reconstruct the lineages of cells in their native context as they organize in tissues, organs, and whole organisms.
