Objective: Low levels of daily energy expenditure (insufficient physical activity and increased sedentary time) have been associated with adverse health outcomes in young people. The Multimedia Activity Recall for Children and Adolescents (MARCA) is a computerized, self-report, use-of-time tool that can assess daily energy expenditure. The study aim was to validate the MARCA for the estimation of energy expenditure in young people, using the criterion standard doubly labeled water.

Materials/methods: Over a 15 day assessment period, 32 participants (10-18 years) completed the MARCA and underwent a doubly labeled water protocol. Indirect calorimetry was used to assess resting metabolic rate. Total daily energy expenditure (TEE) and activity-related energy expenditure (AEE) were estimated from both the MARCA and doubly labeled water. Association and agreement between methods for TEE and AEE were assessed using Spearman correlations and Bland-Altman plots, respectively.

Results: Compared to doubly labeled water, the MARCA over-estimated TEE by an average of 50 kcal/day (limits of agreement -1 589 to 1 490 kcal/day) and under-estimated AEE 105 kcal/day (limits of agreement -1 404 to 1 614 kcal/day). The MARCA showed strong correlation with doubly labeled water for TEE (rho=0.70, p<0.0001) and moderate correlation for AEE (rho=0.56, p=0.0009).

Conclusions: Overall, the MARCA indicated moderate validity for the assessment of daily TEE and AEE. The wide limits of agreement indicate the MARCA has greater utility for group-level rather than individual-level estimates.

Rapid detection of SARS-CoV-2 RNA using a one-step fast multiplex RT-PCR coupled to lateral flow immunoassay.

BMC Infect Dis

December 2024

Laboratory of Molecular Epidemiology and Experimental Pathology, LR16IPT04, Institut Pasteur de Tunis, University of Tunis El Manar, Tunis, LR16IPT04, Tunisia.

Background: The COVID-19 has put emphasis on pivotal needs for diagnosis and surveillance worldwide, with the subsequent shortage of diagnostic reagents and kits. Therefore, it has become strategic for the countries to access diagnostics, expand testing capacity, and develop their own diagnostic capabilities and alternative rapid accurate nucleic acid diagnostics that are at lower costs. Here, we propose a visual SARS-CoV-2 detection using a one-step fast multiplex reverse transcription-PCR (RT-PCR) amplification coupled to lateral flow immunoassay detection on a PCRD device (Abingdon Health, UK).

Determination of microcystins and nodularins in ambient freshwater and seawater by liquid chromatography-mass spectrometry including toxin screening and identification.

Anal Chim Acta

January 2025

HAB Monitoring & Reference Branch, Stressor Detection and Impacts Division, National Centers for Coastal Ocean Science, NOAA National Ocean Service, 331 Fort Johnson Road, Charleston, SC, 29412, USA.

Background: Microcystins (MCs) and nodularins (NODs) produced by cyanobacteria occur in ambient freshwaters and across the freshwater-marine continuum, and pose health threats through drinking and recreational waters, as well as food resources. Approximately 300 MC and NOD toxins have been published, but less than 15 of them are commercially available as toxin standards. Our aim herein was to rapidly identify and quantify all toxin congeners, including those without standards, in water samples even at low abundance by reversed-phase solid phase extraction (SPE)-liquid chromatography-tandem mass spectrometry (LC-MS/MS) to provide insights into toxin levels and potential toxicity.

Water intake in athletes: Agreement between food records and isotope-dilution methods.

Clin Nutr

December 2024

Exercise and Health Laboratory, CIPER, Faculdade de Motricidade Humana, Universidade de Lisboa, Lisboa, Portugal.

Background: Although water intake (WI) is commonly estimated through self-reporting, its inaccuracy is reported. Water turnover (rHO)-derived WI is the reference method; however, it is costly. The study aimed to validate alternative methods for determining WI in a group of athletes.

Article Synopsis
  • Hypocretin deficiency leads to type 1 narcolepsy, which is associated with excessive daytime sleepiness and a high prevalence of overweight and obesity in patients.
  • A study compared energy expenditure in ten males with narcolepsy to nine healthy controls using respiration chamber calorimetry and doubly labelled water.
  • Results showed no significant differences in energy expenditure or physical activity between the two groups, suggesting that weight gain in narcolepsy might be influenced by factors other than metabolism, such as dietary habits.

Research on world-class athletes in endurance events, such as cycling Grand Tours, has reported extreme levels of total energy expenditure. However, it has been argued that over extended periods, such as months, sustained energy expenditure is capped at approximately 2.5 times the basal metabolic rate.
