We previously identified mutants in the rpoE gene of Salmonella enterica serovar Typhimurium (S. Typhimurium) encoding RNA polymerase extracytoplasmic stress response sigma factors σ(E) with altered promoter specificity. The replacement of the conserved R171 residue in the conserved region 4.2 of σ(E) by different amino acid residues exhibited different phenotypes. While R171A almost completely abolished sigma factor activity, R171G and R171C mutant changes imparted a relaxed recognition phenotype to the sigma factor. In the present study, we introduced these mutations into the S. Typhimurium chromosome to investigate their phenotype during ethanol stress and in promoter recognition. Both relaxed sigma factors were found to initiate transcription from a high number of artificial promoters in the S. Typhimurium genome. Both mutants had substantially decreased activity under stress conditions. However, this decreased activity and also the recognition of atypical promoters had no significant effect upon growth, even in stressful conditions.

Our objectives were to use a quantitative literature review to explore dietary and feed factors influencing apparent total-tract digestibility of dry matter (DMD), crude protein (CPD), neutral detergent fiber (NDFD), ether extract (EED), non-structural carbohydrates (NSCD), non-fiber carbohydrates (NFCD), and residual organic matter (rOMD) in equine diets, and to assess their contributions to digestible energy (DE) supplies. Data from 54 studies were modeled using linear mixed-effect regressions, with publication as a random effect to account for study variability. For each nutrient, five models were derived with explanatory variables including: dry matter intake (DMI; % BW/day) and DM (% as-fed), and dietary components (CP, organic matter, EE, NDF, acid detergent fiber, NSC, starch, and NFC as % of DM), and feed types (forage, non-forage fiber, legumes, cereal, and oil proportions).

We propose a hypothesis for the simultaneous emergence of bacteria, archaea, viruses, and mobile elements by sequential and concrete biochemical pathways. The emergence process can be considered analogous to crystallization, where genetic and biochemical systems stabilize as organisms evolve from their common ancestor, the LUCA, which was a non-free-living pool of single operon type genomes including double-stranded (ds) DNA at an ancient submarine alkaline vent. Each dsDNA operon was transcribed by different systems in σ, TFIIB, or TBP genomes.
