Human antibody single domains are a promising new class of antibody fragments. Here we describe methods for the cloning of human V(H) and V(L) genes into phage and phagemid vectors. Furthermore, we provide detailed protocols for the generation of single domain antibody libraries by Kunkel mutagenesis and the analysis of diversity by DNA sequencing and superantigen binding.

Background: Axicabtagene ciloleucel (axi-cel), a chimeric antigen receptor (CAR) T-cell therapy, has significantly improved clinical outcomes in adult patients with relapsed/refractory large B-cell lymphoma (LBCL). Phosphorus (Pi) is an essential nutrient for plants to sustain normal life processes. In this study, we found that the ZmPHO1 proteins had similar molecular weights and the same conserved domain. Phylogenetic and cis-acting element analysis showed that ZmPHO1s were divided into 4 subgroups, in which ZmPHO1;2a and ZmPHO1;2b were closely phylogenetic with OsPHO1;2b, and the promoter region of ZmPHO1s contained abundant abiotic stress-related elements.

Neuroimaging techniques including functional magnetic resonance imaging (fMRI) and electroencephalogram (EEG) have shown promise in detecting functional abnormalities in various brain disorders. However, existing studies often focus on a single domain or modality, neglecting the valuable complementary information offered by multiple domains from both fMRI and EEG, which is crucial for a comprehensive representation of disorder pathology. This limitation poses a challenge in effectively leveraging the synergistic information derived from these modalities.

The outer mitochondrial membrane protein known as mitoNEET was discovered when it was labeled by a photoaffinity derivative of the anti-diabetes medication, pioglitazone. The biological role for mitoNEET and its specific mechanism for achieving this remains an active subject for research. There is accumulating evidence suggesting that mitoNEET could be a component of mitochondrial FeS cofactor biogenesis.
