  • CS/DS proteoglycans are found throughout the body and are involved in various biological functions, including inflammation, wound healing, and the organization of the extracellular matrix.

Article Abstract

The ability of chondroitin/dermatan sulfate (CS/DS) to convey biological information is enriched by the presence of iduronic acid. DS-epimerases 1 and 2 (DS-epi1 and 2), in conjunction with DS-4-O-sulfotransferase 1, are the enzymes responsible for iduronic acid biosynthesis and will be the major focus of this review. CS/DS proteoglycans (CS/DS-PGs) are ubiquitously found in connective tissues, basement membranes, and cell surfaces or are stored intracellularly. Such wide distribution reflects the variety of biological roles in which they are involved, from extracellular matrix organization to regulation of processes such as proliferation, migration, adhesion, and differentiation. They play roles in inflammation, angiogenesis, coagulation, immunity, and wound healing. Such versatility is achieved thanks to their variable composition, both in terms of protein core and the fine structure of the CS/DS chains. Excellent reviews have been published on the collective and individual functions of each CS/DS-PG. This short review presents the biosynthesis and functions of iduronic acid-containing structures, also as revealed by the analysis of the DS-epi1- and 2-deficient mouse models.

The development of long-lasting plasma membrane (PM) and basement membrane (BM) probes is in high demand to advance our understanding of membrane dynamics during differentiation and disease conditions. Herein, we report that the microheterogeneity of heparan sulfate (HS) on fluorescent neo-proteoglycans backbone offers a facile platform for designing membrane probes. Confocal live-cell imaging studies of cancer and normal cell lines with a panel of Cy5 fluorescently tagged neo-proteoglycans confirmed that highly sulfated HS ligands with an l-iduronic acid component (PG@ID-6) induce a prolonged and brighter expression on the PM compared to low-sulfated and uronic acid counterparts.

Extracting parameters such as chemical shifts and coupling constants from proton NMR spectra is often a first step in using spectra for compound identification and structure determination. This can become challenging when scalar couplings between protons are comparable in size to chemical shift differences (strongly coupled), as is often the case with low-field (bench top) spectrometers. Here we explore the potential utility of AI methods, in particular neural networks, for extracting parameters from low-field spectra.

This research work explores the physicochemical characteristics and biological functions of polysaccharides extracted from four selected green seaweed species: Ulva lactuca (UL), Halimeda opuntia (HO), Caulerpa racemosa (CR), and Chaetomorpha antennina (CA). The extracts were investigated for cell-based bio-activities (i.e.

  • TREM2 is an important receptor in the immune system that plays roles in inflammation, cell survival, and neuroprotection, with variations linked to Alzheimer's disease.
  • The study investigates how TREM2 interacts with heparan sulfate (HS) on microglial cells, revealing that TREM2 forms complexes with HS and requires specific structural features of HS for effective binding.
  • Findings suggest that this interaction is crucial for TREM2's role in microglial function, particularly in facilitating the uptake of ApoE3, potentially offering targets for interventions in Alzheimer's disease.

Article Synopsis
  • This study focused on isolating a polysaccharide called Ulva pertusa polysaccharide (UPP) from the Korean seaweed U. pertusa, which shows potential anti-inflammatory effects for bowel diseases.
  • UPP was obtained using hydrothermal extraction and ethanol precipitation, revealing a unique structure that includes specific sulfated sugar compounds linked by glycosidic bonds.
  • Administering UPP in mice with colitis reduced inflammatory factors and improved tight junction proteins, helping to restore a healthy gut environment and suggesting that UPP could be a beneficial treatment for inflammatory bowel disease (IBD).
