Background: Hyalocytes are the main vitreal cell types with critical functions in health and vitreoretinal diseases. Our aim was to develop cultures of human hyalocytes and verify the retention of their initial cellular features after 3 and 6 days of culturing (3 d and 6 d) by analyzing and comparing a few morphological and functional parameters.

Methods: Vitreous samples (n = 22) were collected and vitreous cells and bead-enriched hyalocytes were developed and compared (3 d vs.

Immune drivers of pain resolution and protection.

Nat Immunol

December 2024

F.M. Kirby Neurobiology Center, Boston Children's Hospital, Boston, MA, USA.

Immune cells are involved in the pathogenesis of pain by directly activating or sensitizing nociceptor sensory neurons. However, because the immune system also has the capacity to self-regulate through anti-inflammatory mechanisms that drive the resolution of inflammation, it might promote pain resolution and prevention. Here, we describe how immune cell-derived cytokines can act directly on sensory neurons to inhibit pain hypersensitivity and how immune-derived endogenous opioids promote analgesia.

Article Synopsis
  • * The study found that the lipopolysaccharide (LPS) treatment, which normally promotes M1 polarization, actually increases endogenous MLT secretion in macrophages, indicating a possible protective mechanism.
  • * Results show that LPS activates the TLR4/TRIF pathway leading to MLT secretion, with MLT helping to inhibit M1 polarization and reduce apoptosis in macrophages, highlighting its role in immune response and pathogen clearance.

Article Synopsis
  • * In an experiment with mice, cordycepin treatment significantly reduced signs of colitis, improved overall health, and positively affected gut bacteria composition after a week of administration.
  • * The findings suggest that cordycepin effectively alleviates colitis by promoting a healthy balance between gut microbiota and the immune system's response.
