Solutions containing a high concentration (0.325-0.995 g/ml) of natural-abundance ammonium heptamolybdate tetrahydrate ((NH(4))(6)Mo(7)O(24))·4H(2)O were irradiated at 13 MeV on a proton-beam cyclotron using a standard liquid target. (94m)Tc was produced via the (94)Mo(p,n)(94m)Tc reaction with measured yields of 110±20 MBq for the highest concentrated solution using 5 μA proton beams for 60 min. Saturation yields of up to 40±6 MBq/μA were achieved. Pertechnetate was isolated from the target mixture with 70.9±0.7% efficiency using a solid-phase extraction resin.

The trihydroxamic acid bacterial siderophore desferrioxamine B (DFOB, 1) produced by the DesABCD biosynthetic cluster coordinates metals beyond Fe(iii), which identifies potential to modify this chelator type to broaden metal sequestration and/or delivery applications. Rather than producing discrete chelators by total chemical synthesis from native monomers including -hydroxy--succinyl-cadaverine (HSC, 2), the recombinant siderophore synthetase from CNB-440 (DesD) was used with different substrate combinations to produce biocombinatorial mixtures of hydroxamic acid chelators. The mixtures were screened with Ga(iii) or Zr(iv) as surrogates of immunological positron emission tomography (PET) imaging radiometals Ga(iii) or Zr(iv) to inform known or new coordination chemistry.

Background: Prostate-specific membrane antigen (PSMA) is an ideal target for molecular imaging and targeted radionuclide therapy in prostate cancer. Consequently, various PSMA ligands were developed. Some of these molecules are functionalized with a chelator that can host radiometals, such as Ga for PET imaging.

Tuning aminopolycarboxylate chelators for efficient complexation of trivalent actinides.

Sci Rep

Aqueous Separations and Radiochemistry, Idaho National Laboratory, Idaho Falls, ID, 83415, USA.

The complexation of trivalent lanthanides and minor actinides (Am, Cm, and Cf) by the acyclic aminopolycarboxylate chelators 6,6'-((ethane-1,2-diylbis-((carboxymethyl)azanediyl))bis-(methylene))dipicolinic acid (Hoctapa) and 6,6'-((((4-(1-(2-(2-(2-hydroxyethoxy)ethoxy)ethyl)-1H-1,2,3-triazol-4-yl)pyridine-2,6-diyl)bis-(methylene))bis-((carboxymethyl)azanediyl))bis-(methylene)) dipicolinic acid (Hpypa-peg) were studied using potentiometry, spectroscopy, competitive complexation liquid-liquid extraction, and ab initio molecular dynamics simulations. Two studied reagents are strong multidentate chelators, well-suited for applications seeking radiometal coordination for in-vivo delivery and f-element isolation. The previously reported Hoctapa forms a compact coordination packet, while Hpypa-peg is less sterically constrained due to the presence of central pyridine ring.

We present the production of Co on a small, 13 MeV medical cyclotron utilizing a siphon style liquid target system. Different concentrated iron(III)-nitrate solutions of natural isotopic distribution were irradiated at varying initial pressures and subsequently separated by solid phase extraction chromatography. The radio cobalt (Co and Co) was successfully produced with saturation activities of (0.

Designing and implementing various radionuclide production methods guarantees a sustainable supply, which is important for medical use. The use of medical cyclotrons for radiometal production can increase the availability of gallium-68 (Ga) radiopharmaceuticals. Although generators have greatly influenced the demand for Ga radiopharmaceuticals, the use of medical cyclotrons is currently being explored.
