Towards high throughput metabolic flux analysis in plants. | LitMetric

Research on plant metabolism is currently experiencing the common use of various omics methods creating valuable information on the concentrations of the cell's constituents. However, little is known about in vivo reaction rates, which can be determined by Metabolic Flux Analysis (MFA), a combination of isotope labeling experiments and computer modeling of the metabolic network. Large-scale applications of this method so far have been hampered by tedious procedures of tissue culture, analytics, modeling and simulation. By streamlining the workflow of MFA, the throughput of the method could be significantly increased. We propose strategies for these improvements on various sub-steps which will move flux analysis to the medium-throughput range and closer to established methods such as metabolite profiling. Furthermore, this may enable novel applications of MFA, for example screening plant populations for traits related to the flux phenotype.

Coastal disposal of mine wastes has been recognized as a critical source of metals and metalloids to the water column and seabed sediments, leading to ecotoxicological impacts and benthic habitat loss. Portmán Bay, SE Spain, is one of the most severe cases of coastal impacts due to mine waste dumping. From 1957 to 1990, 57 million tons of metal (Fe, Mn, Zn and Pb) and metalloid-rich (As) artificial soils were discharged into the shoreline, completely filling the bay and extending it seaward.

The high magnetic saturation at flux isolation bridge of interior permanent magnet synchronous motor (IPMSM) enriches the harmonic content of air gap flux density. When IPMSM operates at low speeds, these harmonics of air gap flux density increase torque ripple. An IPMSM with I2V type is proposed to further improve sine degree of no-load air gap flux density, achieving high torque density and low torque ripple.

Sulaibikhat Bay, a shallow estuarine embayment in the southern part of Kuwait Bay, has undergone substantial anthropogenic impacts over the past decade, resulting in altered sediment quality and mercury (T-Hg) contamination. This bay, bordered by the Doha Power Plant and Shuwaikh Port, receives untreated sewage and industrial waste through 17 outfalls, nine of which remain active. To assess the extent of contamination, 44 surface sediment samples were analyzed for T-Hg concentrations and fluxes.

Although nitrifying and N-fixing functional gene microbes play a crucial role in regulating ecosystem nitrogen (N) cycling, understanding of how N and phosphorus (P) fertilization affect their community structure remains limited. N fertilizer levels were set to simulate N deposition and combined with different P fertilizer levels, to explore the soil-root-shoot physiological indexes, and the response of community structure of N-fixing and nitrifying microorganisms that caused changes in the N cycle of the system to fertilization. The results showed that N and P fertilization significantly decreased soil N pool (NH-N, NO-N, IN), the concentrations of shoot TN, root TP, root OC, and root biomass, and slowed down soil N flux (net ammoniation rate, net nitrification rate, and net N mineralization rate).
