Alterations in excitability of low-, medium- and high-threshold single mechanoreceptors (SMRs) of hairless hind paw skin elicited by transcranial electrical stimulation (TES) in analgesic regime were studied in anesthetized rats. Evoked impulsations of low- and medium-threshold SMRs were blocked by TES that lasted for about 1 and 5 min, respectively. TES for 18-20 min in duration abolished the reactions of high threshold SMRs activated only by nociceptive stimuli. Inhibitory effects of TES on all SMR types under study were eliminated by preliminary intraperitoneal or local intracutaneous (near SMRs location) injection of naloxone. Possible mechanisms of analgesic TES effects are discussed. An assumption is made that alterations of the SMR function are connected with an increase of the output of endogenous opioid peptides from the brain during TES.

As transposable elements (TEs) coevolved with the host genome, the host genome exploited TEs as functional regulatory elements of gene expression. Here we show that a subset of KRAB domain-containing zinc-finger proteins (KZFPs), which are highly expressed in mitotically dividing spermatogonia, repress the enhancer function of endogenous retroviruses (ERVs) and that the release from KZFP-mediated repression allows activation of ERV enhancers upon entry into meiosis. This regulatory feature is observed for independently evolved KZFPs and ERVs in mice and humans, suggesting evolutionary conservation in mammals.

Poly(A)-binding protein (Pab1 in yeast) is involved in mRNA decay and translation initiation, but its molecular functions are incompletely understood. We found that auxin-induced degradation of Pab1 reduced bulk mRNA and polysome abundance in WT but not in a mutant lacking the catalytic subunit of decapping enzyme (Dcp2), suggesting that enhanced decapping/degradation is a major driver of reduced translation at limiting Pab1. An increased median poly(A) tail length conferred by Pab1 depletion was likewise not observed in the dcp2Δ mutant, suggesting that mRNA isoforms with shorter tails are preferentially decapped/degraded at limiting Pab1.

Current progress in plant genomics has uncovered important roles of transposable elements (TEs) in gene regulation and has transformed their perception from 'junk DNA' to key genomic players. Recent advances show how stress conditions trigger TE mobilization, introducing new regulatory sequences that can reshape plant responses to environmental changes. This review explores our current knowledge of how TEs, especially those located in gene-rich regions of plant genomes, regulate gene expression at different mechanistic levels.

This study presents the first chromosomal-scale genome assembly of (~626 Mb, 2 = 26), constructed using PacBio HiFi and Hi-C sequencing technologies. Comparative genomic analyses revealed significant recent genomic changes through rapid amplification of transposable elements, particularly long terminal repeat retrotransposons, coinciding with the dramatic climate change during recent uplift of the Hengduan Mountains.
