We find that a common mutation that increases angiotensin I-converting enzyme activity occurs with higher frequency in male patients suffering from refractory temporal lobe epilepsy. However, in their brains, the activity of the enzyme is downregulated. As an explanation, we surprisingly find that carbamazepine, commonly used to treat epilepsy, is an inhibitor of the enzyme, thus providing a direct link between epilepsy and the renin-angiotensin and kallikrein-kinin systems.

Angiotensin-I converting enzyme (ACE) regulates the levels of disparate bioactive peptides, notably converting angiotensin-I to angiotensin-II and degrading amyloid beta. ACE is a heavily glycosylated dimer, containing 4 analogous catalytic sites, and exists in membrane bound and soluble (sACE) forms. ACE inhibition is a frontline, FDA-approved, therapy for cardiovascular diseases yet is associated with significant side effects, including higher rates of lung cancer.

The purpose of this study was to understand the effects of cooking treatment on the protein hydrolysis of beef tripe and the release of potentially bioactive peptides using an in vitro gastrointestinal model. The results showed that digestion promoted the hydrolysis of proteins and release of free amino acids in beef tripe, but cooking treatment significantly reduced them. The sample of the cooked beef tripe after gastrointestinal digestion had the highest antioxidant activity.

Soybean peptide (SP) exhibits significant angiotensin-I-converting enzyme inhibitory (ACEI) activity, however, its strong bitterness restricts its use in food industry. This study aimed to reduce the bitterness of SP by natural deep eutectic solvent (NADES)-driven Maillard reaction (MR). Results showed that both the mixtures of Glucose-NADES and the Glucose-Xylose-NADES formed the hydrogen bonds and shown good thermal stability analyzed by using Fourier transform infrared (FTIR), Differential scanning calorimetry (DSC) and Thermogravimetric analysis (TGA).
