Overdose of pegfilgrastim: case report.

Published: October 2012

Background: Pegfilgrastim is a G-CSF that can be administered in a single dose 24 h after each cycle of chemotherapy.

Case: 59 years-old man with a stage III-B peripheral T lymphoma with CD4 expression. Dose-dense chemotherapy was started with CHOP-14 combined with pegfilgrastim as primary prophylaxis. An error in the dispensing system meant that the patient received pegfilgrastim for 4 consecutive days. He remained under observation in hospital. During the 3 days after the last dose of pegfilgrastim, the patient remained stable, with no remarkable symptoms or associated laboratory abnormalities, and was discharged.

Conclusion: The case we describe and the available literature lead us to suggest close monitoring as the main measure to be adopted when treating a patient for pegfilgrastim overdose.

Introduction: Dose-dense methotrexate, vinblastine, doxorubicin, and cisplatin (ddMVAC) therapy is indicated as first-line or neoadjuvant chemotherapy (NAC) for patients with advanced or metastatic urothelial carcinoma (UC). However, no studies reported ddMVAC therapy with pegfilgrastim (3.6 mg) in Japanese patients.

Literature review analysis of aortitis induced by granulocyte-colony stimulating factor.

Department of Clinical Pharmacy, Weifang People's Hospital, Shandong Second Medical University, Weifang, China.

Background: Recombinant human granulocyte-colony stimulating factors (G-CSF)-induced aortitis is a rare but particularly serious adverse event, commonly seen in cancer patients undergoing chemotherapy. The aim of this article is to clarify the clinical characteristics of G-CSF- induced aortitis and provide effective references for clinical diagnosis and intervention.

Methods: Case reports of adverse reactions of aortitis induced by G-CSF were collected from the relevant databases.

Pegfilgrastim, a 40 kDa PEGylated form of recombinant human granulocyte colony-stimulating factor (rhG-CSF), is a biotherapeutic protein used to treat chemotherapy-induced neutropenia. To ensure the product is safe and effective, stringent monitoring of product-related impurities, particularly those arising from oxidative degradation, is necessary. This study focuses on the isolation and characterization of oxidized variants in pegfilgrastim using a multi-step approach that includes method transfer to semi-preparative High-Performance Liquid Chromatography (HPLC), mass spectrometry, and an in vitro cell-based potency assay (CBPA).

Epiglottitis is a critical infection that can result in upper airway obstruction. While bacterial infections are the most common cause of acute epiglottitis, Candida epiglottitis remains relatively rare. We report a case involving an 82-year-old male undergoing chemotherapy for small cell lung cancer.

Chronic g-CSF increases the neutrophil-lymphocyte ratio and decreases plasma corticosterone concentrations in Peromyscus maniculatus without impacting compulsive-like stereotypy.

Article Synopsis
  • The study examines the link between brain-immune mechanisms and neurocognitive rigidity in deer mice, particularly focusing on rigid motor stereotypy and systemic inflammation markers like the neutrophil-lymphocyte ratio (NLR).
  • Researchers administered a recombinant human granulocyte colony-stimulating factor (g-CSF) to increase NLR and observed its effects on stereotypical behavior, NLR, and plasma corticosterone levels.
  • Results indicated that NLR and plasma corticosterone levels did not predict stereotypical behavior, and while chronic g-CSF treatment raised NLR and lowered corticosterone, it did not affect stereotypical expressions; more research is needed to understand the potential immune-inflammation role in spontaneous stereotypy.
