Single band coherent anti-Stokes Raman scattering (CARS) microscopy is one of the fastest implementation of nonlinear vibrational imaging allowing for video-rate image acquisition of tissue. This is due to the large Raman signal in the C-H-stretching region. However, the chemical specificity of such images is conventionally assumed to be low. Nonetheless, CARS imaging within the C-H-stretching region enables detection of single cells and nuclei, which allows for histopathologic grading of tissue. Relevant information such as nucleus to cytoplasm ratio, cell density, nucleus size and shape is extracted from CARS images by innovative image processing procedures. In this contribution CARS image contrast within the C-H-stretching region is interpreted by direct comparison with Raman imaging and correlated to the tissue composition justifying the use of CARS imaging in this wavenumber region for biomedical applications.

Osteoarthritis (OA) is a complex disease of cartilage characterised by joint pain, functional limitation, and reduced quality of life with affected joint movement leading to pain and limited mobility. Current methods to diagnose OA are predominantly limited to X-ray, MRI and invasive joint fluid analysis, all of which lack chemical or molecular specificity and are limited to detection of the disease at later stages. A rapid minimally invasive and non-destructive approach to disease diagnosis is a critical unmet need.

Co(CH) ( = 1-6) complexes produced with laser vaporization in a supersonic molecular beam are studied with infrared photodissociation spectroscopy and computational chemistry. Infrared spectra are measured in the C-H stretching region using the method of tagging with argon atoms to enhance the photodissociation yields. C-H stretch vibrations for all clusters studied are shifted to lower frequencies than those of the well-known acetylene vibrations from ligand → metal charge transfer interactions.

Stimulated Raman scattering (SRS) microscopy is a powerful vibrational imaging technique with high chemical specificity. However, the insufficient tuning range or speed of light sources limits the spectral range of SRS imaging and, hence, the ability to identify molecular species. Here, we present a widely tunable fiber optical parametric oscillator with a tuning range of 1470 cm, which can be synchronized with a Ti:sapphire laser.

Breast cancer, a leading cause of female mortality due to delayed detection owing to asymptomatic nature and limited early diagnostic tools, was investigated using a multi-modal approach. Plasma-derived small EVs from breast cancer patients (BrCa, n = 74) and healthy controls (HC, n = 30) were analyzed. We investigated the interlinked N-H and N-C photochemistry of primary and secondary amines via the state-resolved detection of vibrationally excited CH product and H atom product by 200-235 nm dimethylamine photodissociation using resonance-enhanced multiphoton ionization (REMPI) and velocity map imaging (VMI) techniques. The out-of-plane bending (ν) vibrationally excited CH showed a bimodal translational energy distribution that became unimodal with a near-zero product yield at longer photolysis wavelengths (λ). In contrast, a unimodal distribution was observed for the C-H stretching (ν) vibrationally excited CH products with an almost constant product yield in the examined λ region.
