The aim of this study was to develop a sustained release converse thermosensitive hydrogel for intra-articular injection using chitosan-glycerol-borax as matrix, its physical properties and biocompatibility were investigated. Taking gelation time and gelation condition as index, the influence of concentration of chitosan, ratio of chitosan to glycerol, pH on physical properties of hydrogel were investigated. And then the in vitro drug release, rheological properties and biocompatibility were studied. The thermosensitive hydrogel flows easily at room temperature and turns to gelation at body temperature, which can certainly prolong the release of drug and has good biocompatibility.

Topical transdermal drug delivery for psoriasis remains a challenge because of the poor solubility of hydrophobic drugs and the limited penetration of the stratum corneum. In this study, a near-infrared (NIR) light-responsive thermosensitive hydrogel (PDLLA-PEG-PDLLA, PLEL)-based drug reservoir is developed that directly incorporated gold nanorods (GNRs) and methotrexate (MTX) in the sol state at low temperature, which is referred to as PLEL@GNR+MTX. The in vitro anti-psoriasis experiment indicated that, GNRs, as photothermal cores of composite hydrogel, not only triggered keratinocyte apoptosis but also promoted MTX release in a synergistic manner.

To design a multifunctional nanozyme hydrogel with antibacterial, photo-responsive nitric oxide-releasing, and antioxidative properties for promoting the healing of infected wounds. We first developed ultra-small silver nanoparticles (NPs)-decorated sodium nitroprusside-doped Prussian blue (SNPB) NPs, referred to as SNPB@Ag NPs, which served as a multifunctional nanozyme. Subsequently, this nanozyme, together with geniposide (GE), was incorporated into a thermo-sensitive hydrogel, formulated from Poloxamer 407 and carboxymethyl chitosan, creating a novel antibacterial wound dressing designated as GE/SNPB@Ag hydrogel.
