Resveratrol, piceatannol, ε-viniferin, r-viniferin, r2-viniferin, and hopeaphenol are naturally occurring polyphenols, associated with potentially beneficial health effects. We developed a rapid liquid chromatography-ultraviolet detection (LC-UV) method, allowing for the simultaneous determination of these six compounds in biological samples in less than 2.5 min with standard LC equipment. Using this method for the assessment of the stability of the six analytes, we demonstrated that all stilbene polyphenols disappear rapidly in Dulbecco's modified Eagle's medium (e.g., half-life of resveratrol of 1 h). In contrast, the tetramer hopeaphenol was stable over the maximum incubation time of 72 h. In incubations with liver microsomes, ε-viniferin was rapidly glucuronidated, although to a lower extent than resveratrol. Hopeaphenol was not glucuronidated at all. Given that glucuronidation is the major metabolic pathway for polyphenols, hopeaphenol might exhibit significantly different pharmacokinetic properties than other polyphenols. When chemical and metabolic stability as well as biological activity of hopeaphenol are taken together, these findings warrant further investigation of this polyphenol.

Clozapine and its metabolites require close therapeutic monitoring (TDM) in patients due to poor correlation between the administrated doses and resulting plasma concentrations, the narrow therapeutic interval, high inter-individual variability, and the risk of serious side effects once toxic levels are exceeded. The aim of the study was to develop a simple (relatively cheap) LC-UV method for the quantification of clozapine and its metabolites in plasma and urine samples. For sample preparation, liquid-liquid extraction (LLE) in -octanol was more efficient and less limiting in injection volumes compared to the in-situ formation of SUPRAS.

A new method is described for acetone (C(CH)O) determination in water samples. The method is based on the reaction with 4-(dimethylamino)benzaldehyde (DMAB) in dimethyl sulfoxide (DMSO) in slightly basic medium, resulting in a highly fluorescent compound with fluorescent wavelengths undisturbed by other common fluorescent compounds. Because of their topical application in patients and meaningful UV/VIS absorptive properties, the degradation and potential toxicity under irradiation of diflunisal (DIF) and naphazoline (NAF) were studied. In addition, the impact of pH on their photostability was examined, showing the highest degradation of acidic DIF at pH 1 and 13 and the highest degradation of basic NAF at pH below 7. An LC-UV analysis and chemical tests showed the first-order kinetics for their degradation and generation of reactive oxygen species (ROS).
