Introduction: The quality of Coleus forskohlii is often evaluated by high performance liquid chromatography (HPLC), using bioactive labdane diterpenoids as chemical markers. However, the existing sample preparation methods for the analysis of diterpenoids in C. forskohlii are generally labour-intensive, time-consuming and require large volumes of solvents.

Objective: To establish an efficient matrix solid-phase dispersion (MSPD) extraction method for the simultaneous analysis of five bioactive diterpenoids in C. forskohlii by HPLC.

Methodology: Herbal samples were prepared by an optimised MSPD procedure using C(18) as the sorbent. The quantification of the diterpenoids was achieved by HPLC with evaporative light scattering detector (ELSD), and the identification of the five compounds was performed by HPLC with tandem mass detector (MS/MS). The efficiency of the MSPD method was also compared with other extraction techniques including Soxhlet extraction, heat reflux extraction, ultrasonic-assisted extraction and microwave-assisted extraction.

Results: The MSPD extracted five diterpenoids with satisfactory recoveries ranging from 98.36% to 102.08%. Compared with other extraction methods, the proposed MSPD method had the advantages of combining extraction and clean-up into a single step, consuming less time and requiring lower solvent volumes.

Conclusion: The MSPD method is simple, rapid and efficient for the extraction of labdane diterpenoids from C. forskohlii. The MSPD procedure coupled with HPLC-ELSD or HPLC-MS/MS is suitable for the quantification and identification of the diterpenoids in C. forskohlii.

This review highlights the therapeutic potential of epigallocatechin gallate (EGCG) and forskolin in managing retinal diseases, with a focus on glaucoma, age-related macular degeneration (AMD), and diabetic retinopathy. EGCG, a potent polyphenol from green tea, exhibits significant antioxidant, anti-inflammatory, and neuroprotective effects, making it a promising candidate for reducing oxidative stress and inflammation in ocular tissues. Forskolin, a diterpene from Coleus forskohlii, increases cyclic AMP (cAMP) levels, which helps lower intraocular pressure (IOP) and provides neuroprotection.

Royal Botanic Gardens Kew, Richmond, London, TW9 3AE, UK.

The Plectranthinae clade, which includes genera such as Plectranthus, Ocimum, and Aeollanthus, is well known for its diverse array of diterpenoids. While numerous studies have deepened the understanding of diterpene diversity across the clade, Aeollanthus species remain underexplored, with only two studies focusing on their diterpene profiles. The NMR-based chemical profiling of the EtOAc leaf extract of the rocky and succulent species Aeollanthus buchnerianus Briq.

Instituto de Química Orgánica General (IQOG-CSIC), Juan de la Cierva 3, 28006 Madrid, Spain. Electronic address:

Article Synopsis
  • Coleus forskohlii supplements (FKSs), popular for weight management due to forskolin, face issues like fraud and adulteration.
  • A new method using Flow Injection Analysis-Mass Spectrometry (FIA-MS) was developed to effectively detect these issues in FKSs.
  • The method revealed that 45% of tested samples did not contain forskolin, and there was a 69% discrepancy between the actual and labeled forskolin content, demonstrating FIA-MS as a reliable technique for quality assessment.

November 2024

Department of Agricultural Biotechnology, Bidhan Chandra Krishi Viswavidyalaya, Nadia, Mohanpur, West Bengal, 741252, India.

Article Synopsis
  • Forskolin, derived from the roots of Coleus forskohlii, has therapeutic potential and this study aimed to develop a reliable method for regenerating these plants year-round for consistent forskolin production.
  • The researchers found that using 1.5 mg/L meta-topolin increased shoot and leaf output significantly, with optimal results achieved through a combination with auxin.
  • Results showed high plant survival rates during acclimatization, confirmed genetic fidelity with the mother plant, and a notable increase in forskolin levels in the in vitro roots, suggesting this method could support commercial production and conservation efforts.
