Pharmaceutical hydrates have been used as clinical development candidates and in marketed products. The physical stability of hydrates can pose unique challenges to their development because of their particular sensitivity to the moisture levels in their surroundings. By conducting simple experiments early during the form selection phase of a drug candidate's development, a basic understanding of the thermodynamic and kinetic aspects of a hydrate form's stability can be attained that can facilitate the successful navigation of these challenges. Differential scanning calorimetry was used to determine the thermal and kinetic properties of a number of pharmaceutically relevant hydrates. The activation energy (E(a)) of dehydration and dehydration onset temperature (T(onset)) of survey compounds were compiled and analyzed. A significant number of compounds possessed both high E(a) and high T(onset) of dehydration, suggesting that these hydrate crystal forms were particularly stable. The results of these studies suggest that dehydration E(a) and dehydration T(onset) together can be used as early indicators of a crystalline hydrate's physical stability and can alert to potential challenges in developing hydrate crystal forms of drug candidates.

The development of stable biopharmaceutical formulations, such as monoclonal antibodies, poses a great challenge in the pharmaceutical industry. This study investigated the stabilizing effect of 2-hydroxypropyl-β-cyclodextrin (HP-β-CD) in liquid and solid formulations of infliximab during processing and storage. The solid formulation was produced by a scaled-up high-speed electrospinning method, resulting in a product suitable for reconstitution with excellent dissolution properties.

Perovskite technologies has taken giant steps on its advances in only a decade time, from fundamental science to device engineering. The possibility to exploit this technology on a thin flexible substrate gives an unbeatable power to weight ratio compares to similar photovoltaic systems, opening new possibilities and new integration concepts, going from building integrated and applied photovoltaics (BIPV, BAPV) to internet of things (IoT). Metal-free molecular perovskites have shown great potential for X-ray detection due to their tunable chemical structures, low toxicity, and excellent photophysical properties. The local electric field (LEF) plays an important role in the catalytic process; however, the precise construction and manipulation of the electric field microenvironment around the active site remains a significant challenge. Here, we have developed a supramolecular strategy for the implementation of a LEF by introducing the host macrocycle 18-crown-6 (18C6) into a cobalt phthalocyanine (CoPc)-containing covalent organic framework (COF). Utilizing the supramolecular interaction between 18C6 and potassium ion (K), a locally enhanced K concentration around CoPc can be built to generate a LEF microenvironment around the catalytically active Co site.
