Article Synopsis
  • Condyloma acuminatum is usually caused by HPV and typically appears as pale, raised lesions, but some patients displayed pigmented lesions resembling seborrhoeic keratoses, which were confirmed to be HPV-positive.
  • In a retrospective study, 31 patients were analyzed for these unusual pigmented lesions, with most presenting as slow-growing and located at the base of the penis or pubis, primarily exhibiting the HPV-6 genotype.
  • The study concludes that these 'seborrhoeic keratosis-like' condylomas share clinical characteristics with seborrhoeic keratoses but are distinctly linked to HPV, with only a limited number of similar cases documented in existing literature.

Article Synopsis
  • Proliferative verrucous leukoplakia (PVL) is distinct from other oral leukoplakias (OLs) in its clinical behavior, and this study aimed to identify molecular differences through DNA methylation analysis.
  • A genome-wide analysis revealed 1,815 differentially methylated CpGs between PVL and homogeneous leucoplakia (HL), with HL showing hypermethylation and involvement of genes related to cancer prognosis.
  • The research developed a logistic regression model that can differentiate between PVL, HL, and healthy samples, highlighting the potential for using methylation patterns as diagnostic biomarkers for oral lesions.

Molecular landscape of proliferative verrucous leukoplakia: a systematic review.

Br J Oral Maxillofac Surg

February 2024

Cancer Sciences, Faculty of Medicine, University of Southampton, Southampton, UK; Institute for Life Sciences, University of Southampton, Southampton, UK.

Article Synopsis
  • Proliferative verrucous leukoplakia (PVL) is a rare oral condition with a potential to evolve into oral cancers, lacking predictive biomarkers for its progression.
  • A review of 685 cases revealed PVL’s genomic profile includes initial high loss of heterozygosity (LOH) and low copy number alterations, with later mutations found in specific genes but no TP53 mutations.
  • Differences in LOH and specific chromosomal deletions and amplifications suggest similarities with oral verrucous carcinoma (OVC) and oral squamous cell carcinoma (OSCC), indicating a need for further research to clarify PVL's genomic landscape.

Squamous cell carcinoma (SCC) is the most common malignant tumour of the penis. The 2022 WHO classification reinforces the 2016 classification and subclassifies precursor lesions and tumours into human papillomavirus (HPV)-associated and HPV-independent types. HPV-associated penile intraepithelial neoplasia (PeIN) is a precursor lesion of invasive HPV- associated SCC, whereas differentiated PeIN is a precursor lesion of HPV-independent SCC.
