Synthetic biology and genome-scale protein work both require rapid and efficient cloning, expression and purification. Tools for co-expression of multiple proteins and production of fusion proteins with purification and solubility tags are often desirable. Here we present a survey of plasmid vectors that provide for some of these features with a focus on tools for rapid cloning and traceless tagging - a setup that facilitates removal of fusion tags post-purification leaving behind no 'scar' on the final construct. Key features are reviewed, including plasmid replication origins and resistance markers, transcriptional promoters, cloning methods, and fusion tags and their removal by proteolysis. We describe a vector system called pHLIC, which assembles features for simple cloning, overexpression, facile purification, and traceless cleavage, as well as flexibility in modifying the vector to exchange fusion tags.

Transpeptidases are specialized enzymes that have evolved for site-selective modification of peptides and proteins at their backbone termini. Approaches for adapting transpeptidases to catalyze side chain modifications are substantially more restricted, and typically rely on large recognition tags or require specific reaction conditions that are not easily compatible with broader applications. Here we show that the engineered asparaginyl ligase AEP1 catalyzes direct isopeptide ligation by accepting an internal 2,3-diaminopropionic acid (Dap) residue adjacent to Leu, a motif that mimics the canonical N-terminal Gly-Leu substrate.

The synthesis of chiral amines is of critical importance but still challenging. Here, we present a self-sufficient and reusable dual-enzyme nanoreactor for chiral amine synthesis, featuring Z-mediated site-specific immobilization of amine dehydrogenase (AmDH) and glucose dehydrogenase (GDH) onto mesoporous silica nanoflowers (MSN). Molecular dynamics simulations revealed that the Z tag was bound to MSN via electrostatic interactions, thus maintaining the fusion enzyme's active pocket accessibility and improving its catalytic performance.
